Chronic myeloid leukemia (CML) is a myeloproliferative disease characterized by the formation of BCR-ABL (breakpoint cluster region-Abelson) oncoprotein. As many patients display therapeutic resistance, development new drugs based on semisynthetic products represents potential approach for treating disease. In this study, we investigated cytotoxic activity, possible mechanism action hybrid compound betulinic acid (BA) and brosimine B in CML cell lines that are sensitive (K-562) resistant (K-562R) to imatinib, addition evaluating lower doses imatinib combination with compound. The effects compound, its apoptosis, cycle, autophagy oxidative stress were determined. was K-562 (23.57 ± 2.87 μM) K-562R (25.80 3.21 cells, synergistic effect observed when it associated imatinib. Apoptosis mediated caspase 3 9 intrinsic pathway, cycle evaluation showed arrest at G0/G1. addition, increased production reactive oxygen species induced increasing LC3II Beclin-1 mRNA levels. Results suggest causes death both imatinib-sensitive -resistant may hold as anticancer treatment against CML.

OBJECTIVES: Betulinic acid is pentacyclic triterpenoid known to exert antitumor effects by modulating many cellular pathways in various human malignancies.However, its modulatory role autophagy renal cell carcinoma remains unclear.Here, we observed how betulinic affects cells.METHODS: After treating cells with acid, determined the gene expression and protein levels of Beclin-1 ATG-5 qPCR ELISA assay observe on autophagy.RESULTS: The results demonstrated that level was low untreated metastatic adenocarcinoma ACHN increased response 25 μM 50 treatment.ATG-5 decreased primary clear CAKI-2 treated acid.In results, caused a decrease at concentration cells.CONCLUSION: In current study, it concluded may differ depending their origin these vary accordingly (Fig. 4, Ref. 40).
