Maintaining mitochondrial homeostasis is a potential therapeutic strategy for various diseases, including neurodegenerative cardiovascular metabolic disorders, and cancer. Selective degradation of mitochondria by autophagy (mitophagy) fundamental quality control mechanism conserved from yeast to humans. Indeed, small-molecule modulators mitophagy are valuable pharmaceutical tools that can be used dissect complex biological processes turn them into drugs. In the past few years, pharmacological regulation has shown promising efficacy in disease models. However, with increasing number chemical modulator studies, frequent methodological flaws observed, leading some studies draw unreliable or misleading conclusions. This review attempts (a) summarize molecular mechanisms mitophagy; (b) propose Mitophagy Modulator Characterization System (MMCS); (c) perform comprehensive analysis methods characterize modulators, covering publications over 20 years; (d) provide novel targets intervention mitophagy. We believe this will panorama current research on promote development safe robust introducing high standards.

Polydatin (PD), a natural product derived from Polygonum cuspidatum, has anti-inflammatory and antioxidant effects significant benefits in treating allergic diseases. However, its role mechanism rhinitis (AR) have not been fully elucidated. Herein, we investigated the effect of PD AR. AR model was established mice with OVA. Human nasal epithelial cells (HNEpCs) were stimulated IL-13. HNEpCs also treated an inhibitor mitochondrial division or transfected siRNA. The levels IgE cellular inflammatory factors examined by enzyme linked immunosorbent assay flow cytometry. expressions PINK1, Parkin, P62, LC3B, NLRP3 inflammasome proteins, apoptosis proteins tissues measured Western blot. We found that suppressed OVA-induced thickening eosinophil accumulation mucosa, reduced IL-4 production NALF, regulated Th1/Th2 balance. In addition, mitophagy induced after OVA challenge IL-13 stimulation. Meanwhile, enhanced PINK1-Parkin-mediated but decreased reactive oxygen species (mtROS) production, activation, apoptosis. PD-induced abrogated PINK1 knockdown Mdivi-1 treatment, indicating key PINK1-Parkin mitophagy. Moreover, damage, mtROS under exposure more severe treatment. Conclusively, may exert protective on promoting mitophagy, which further suppresses tissue damage through decreasing activation.
