ABSTRACT Alzheimer´s disease (AD) is the most frequent neurodegenerative disorder in world and characterised by loss of memory other cognitive functions. Metabolic changes associated with AD are important players development disease. However, mechanism underlying these still unknown. Extracellular vesicles (EVs) nano‐sized particles that play an role regulating pathophysiological processes a non‐invasive manner to obtain information cell secreting them. The analysis brain‐derived EVs (bdEVs) will provide new insights metabolic AD. To characterize bdEVs AD, we optimised method isolate them from tissue different brain regions, obtaining highest enrichment isolations temporal cortex. We performed unbiased untargeted metabolomics on post‐mortem human cortex same region patients healthy controls. Both, univariate multivariate statistical were used determine metabolites influence separation between Interestingly, clear control groups was obtained bdEVs, which allowed select 12 relevant features validated PLS‐DA model. Furthermore, comparison identified 68 common features. pathway showed alanine, aspartate glutamate arginine, phenylalanine, tyrosine significant ones tryptophan pathway, had very high groups, albeit not significant. Exosomes are extracellular vesicles secreted by cells and rich in genetic material proteins. The surfaces of exosome membranes contain many blast-specific markers that provide an important basis for disease diagnosis, progression, treatment. Herein, we searched the Web Science core collection (SCI-EXPENED) database research review articles on "exosomes" "biomarkers" or "diagnostics" "liquid biopsy" as topics between 2010 2024. Bibliometric analysis revealed exosomes have received increasing levels attention biomarkers, with China contributing most to these studies. focus marker diagnoses cancer, inflammation, diabetes, well neurodegenerative cardiovascular diseases. Chromatography, mass spectrometry, Raman spectroscopy, other techniques typically used analyze exosomal nucleic acids, proteins, metabolites, commonly test samples including plasma, serum, urine, saliva, cerebrospinal fluid, bodily fluids. Research into tumor has mainly focused eight highly prevalent cancers, lung, breast, prostate cancers. This paper focuses biomarkers uses a bibliometric tool system use their contents diagnosis field 2024, analyzes development prospects, discusses future exosome-mediated efforts diagnosing treating diseases, is expected reference studying applying markers.

As an emerging source for liquid biopsy, exosomes hold significant promise clinical diagnosis. However, commonly used exosome isolation methods (e.g., ultracentrifugation) suffer from low throughput a large number of samples. Herein, dysprosium–metal organic framework was synthesized and doped with nanofibers by electrospinning efficient capture body fluid. With the integration multichannel pipet or robot automatic workstation, high can be achieved samples reproducibility. To evaluate value developed method, urinary were enriched 34 liver disease different stages profiling metabolites mass spectrometry. The results showed that HCC, cirrhosis, healthy controls significantly differentiated Random Forest classification model. has promising applications in exosome-based biopsy large-scale
