Interleukin-12 (IL-12) is considered to be a promising cytokine for enhancing an antitumor immune response; however, recombinant IL-12 has shown significant toxicity and limited efficacy in early clinical trials. Recently, many strategies delivering tumor tissues have been developed, such as modifying IL-12, utilizing viral vectors, non-viral cellular vectors. Previous studies found that the fusion of with extracellular matrix proteins, collagen, factors way enhance its therapeutic potential. In addition, demonstrated vectors are good platform, variety viruses oncolytic viruses, adenoviruses, poxviruses used deliver IL-12—with testing previously conducted various cancer models. The local expression tumors based on delivery avoids systemic while inducing effective immunity acting synergistically other therapies without compromising safety. lipid nanoparticles currently most mature drug system. Moreover, cells also carriers because they can effectively substances tumors. this article, we will systematically discuss anti-tumor effects own or combination different strategies.

Antiangiogenic medications for cancer treatment have generally failed in showing substantial benefits terms of prolonging life on their own; effects are noticeable only when combined with chemotherapy. Moreover, treatments based prolonged antiangiogenics administration demonstrated to be ineffective stopping tumor progression. In this scenario, nanotherapeutics can address certain issues linked existing antiangiogenic treatments. More specifically, they provide the ability target tumor's blood vessels enhance drug accumulation and manage release, ultimately decreasing undesired side effects. Additionally, enable multiple angiogenesis inhibitors at same time as Key reports field include design polymeric nanoparticles, inorganic vesicles, hydrogels loading substances like endostatin interleukin-12. Furthermore, nanoformulations been proposed efficiently control relevant pro-angiogenic pathways such VEGF, Tie2/Angiopoietin-1, HIF-1α/HIF-2α, TGF-β, providing powerful approaches block growth metastasis. article, we outline a selection that developed past ten years.

In situ vaccination (ISV) has emerged as a promising strategy in cancer immunotherapy, offering targeted approach that uses the tumor microenvironment (TME) to stimulate an immune response directly at site. This method minimizes systemic exposure while maintaining therapeutic efficacy and enhancing safety. Recent advances nanotechnology have enabled new approaches ISV by utilizing nanomaterials with unique properties, including enhanced permeability, retention, controlled drug release. employing can induce immunogenic cell death reverse immunosuppressive hypoxic TME, thereby converting "cold" into "hot" facilitating more robust response. review examines mechanisms through which nanomaterials-based enhances anti-tumor immunity, summarizes clinical applications of these strategies, evaluates its capacity serve neoadjuvant therapy for eliminating micrometastases early-stage patients. Challenges associated translation ISV, nanomaterial metabolism, optimization treatment protocols, integration other therapies such radiotherapy, chemotherapy, photothermal therapy, are also discussed. Advances immunotherapy continue expand possible potentially leading improved outcomes across broad range types.

Dictyophora indusiata is commonly utilized as a functional food in China and other Asian countries. The peach-shaped phase of this fungus nutritionally taste-wise similar to its mature fruiting bodies. However, there limited research on the polysaccharides found D. indusiata. A heteropolysaccharide was extracted from volva (DIVP). Analyses using high-performance gel permeation chromatography, methylation NMR revealed that DIVP comprises glucose, glucuronic acid, galactose, mannose. Its structure features backbone consists →3)-β-D-Glcp-(1→ units with branches at →4)-β-D-Glcp-(1→, →6)-α-D-Galp-(1→ terminal α-Manp-(1→ residues. Physicochemical assessments including X-ray diffraction, thermal, zeta potential viscosity characterization indicated semi-crystalline polymer exhibiting excellent physical thermal stability. Cytokine antibody array proteome profiler human phosphokinase analyses demonstrated downregulates expression levels cytokines alters phosphorylation status 16 proteins U937 macrophages induced by lipopolysaccharides, indicating anti-inflammatory activity. These findings suggest polysaccharide primarily composed β-1,3-glucan, which exhibits stable physicochemical properties activity, providing foundation for use an agent or food.
