Bone metastasis is a common cause of death in patients with non-small cell lung cancer (NSCLC), approximately 30-40% NSCLC eventually developing bone metastases. metastasis, especially the occurrence skeletal-related events (SREs), significantly reduces overall survival (OS) and quality life (QoL) patients. Although bone-targeting agents (BTAs) have been shown to reduce SREs improve QoL metastases, prognosis for these remains poor. Understanding underlying molecular pathways crucial development novel therapeutic approaches. complex, multistep process that involves interactions between tumor cells microenvironment. The microenvironment provides fertile soil cells, crosstalk among various signaling secreted factors also plays role regulating progression NSCLC. Arsenic Nanoparticles Trigger Apoptosis via Anoikis Induction in OECM-1 Cells DOI Open Access

Arsenic compounds have been used as therapeutic alternatives for several diseases including cancer. In the following work, we obtained arsenic nanoparticles (AsNPs) produced by an anaerobic bacterium from Salar de Ascotán, in northern Chile, and evaluated their effects on human oral squamous carcinoma cell line OECM-1. Resazurin reduction assays were carried out these cells using 1–100 µM of AsNPs, finding a concentration-dependent viability that was not observed non-tumoral gastric mucosa-derived GES-1. To establish if associated with apoptosis induction, markers like Bcl2, Bax, cleaved caspase 3 analyzed via Western blot, executor caspases 3/7 luminometry, DNA fragmentation TUNEL assay, 100 cisplatin positive control. OECM-1 treated AsNPs showed induction both extrinsic intrinsic apoptotic pathways, which can be explained significant decrease P-Akt/Akt P-ERK/ERK relative protein ratios, increase PTEN p53 mRNA levels Bit-1 levels. These results suggest prospective mechanism action involves potential interaction extracellular matrix (ECM) components reduces attachment subsequently triggers anoikis, anchorage-dependent type apoptosis.

Tumor metastasis is responsible for 90% of cancer-associated deaths, and its early detection may decrease the likelihood mortality. Studies have demonstrated that results from interaction between "seeds" (tumor cells) "soil" (pre-metastatic niche, PMN). As first most abundant immune cells to be recruited PMN, neutrophils play a key role in ultimate formation metastatic foci through mechanisms such as supporting tumor cell growth, promoting angiogenesis, shaping an immune-suppressive microenvironment. In this study, two distinct types sialic acid (SA)-modified liposomes were prepared target regulate pro-metastatic l-selectin receptor. One these liposomes, named ICG@SAL, was used encapsulate indocyanine green (ICG) specifically designed cancer metastasis. The other liposome, referred ABE/Cur@SAL, co-loaded abemaciclib (ABE) curcumin (Cur), with intention suppressing progression tumor. Fluorescence imaging mouse spontaneous model indicated ICG@SAL faster targeting stronger accumulation organs than unmodified ICG (ICG@CL). This suggested could detect at stage. therapy experimental lung ABE/Cur@SAL inhibit regulatory T (Treg) proliferation, enhance effector activity reduce tumorigenic factor release, implying Overall, our work provided sensitive convenient approach diagnosis treatment employed metastasis, while development when identified.
