Metagenomic next-generation sequencing (mNGS) allows untargeted identification of a broad range pathogens, including rare or novel microorganisms. Despite the recognition mNGS as valuable diagnostic tool for infections, most relevant indications this innovative strategy remain poorly defined. We aimed to assess determinants positivity and clinical utility mNGS.

Abstract Background As sequencing is becoming more broadly available, virus discovery continues. Small DNA viruses contribute to up 60% of the overall load in pigs. Porcine circoviruses (PCVs) are small with a single‐stranded circular genome. They common pig breeds and have not been properly addressed for their potential risk xenotransplantation. Whereas PCV1 non‐pathogenic pigs, PCV2 has associated various disease manifestations. Recently two new described, PCV3 PCV4. The family Circoviridae includes the genera Circovirus and Cyclovirus. Cycloviruses have been found in serum samples from chronic HBV, HCV, or HIV-infected individuals as well asymptomatic blood donors. However, research on cycloviruses is relatively limited. We used viral metagenomics to mine, analyze, visualize human virome, successfully identifying three new genomes, each encoding Rep Capsid proteins. These proteins are crucial for replication host-cell interaction: protein involved initiating genome replication, while plays a key role assembly of virions virus's ability interact with host immune systems. Distance matrix phylogenetic analyses show that these share high sequence similarity viruses both humans animals across different regions Africa. This finding not only confirms presence previously uncharacterized blood, but also provides insight into their potential transmission ecological significance. Further needed explore functional roles pathogenesis, particularly how they may influence immunity contribute infections. Additionally, studies investigating range mechanisms cross-species will be essential understanding broader implications animal health.

SUMMARY Xenotransplantation, the transplantation of living organs, tissues, or cells between species, carries potential to address global shortage human organs for patients with end-stage organ failure. Recent advances in genetic engineering have improved prospects clinical xenotransplantation by reducing immune and inflammatory responses grafts, controlling coagulation on endothelial surfaces, modifying viral risks, including porcine endogenous retrovirus (PERV). Management infectious risks posed requires meticulous attention biosecure breeding microbiological surveillance source animals recipients consideration novel infection control requirements. Summary Enteric viruses are the main cause of acute gastroenteritis worldwide with a significant morbidity and mortality, especially among children aged adults. Some enteric also disseminated infections severe neurological manifestations such as poliomyelitis. Protective immunity against these is not well understood in humans, most knowledge coming from animal models, although development poliovirus rotavirus vaccines has extended our knowledge. In classical view, innate involves recognition foreign DNA or RNA by pathogen receptors leading to production interferons other inflammatory cytokines. Antigen uptake presentation T cells B then activate adaptive and, case mucosal immunity, induce secretion dimeric IgA, more potent immunoglobulins viral neutralization. The study Inborn errors (IEIs) offers natural opportunity nonredundant toward pathogens. viruses, patients defective antibodies at risk developing complications. Moreover, recent description low absent antibody protracted associated hepatitis reinforces prominent role control virus.
