ABSTRACT Exoskeletons are a defining character of all arthropods that provide physical support for their segmented bodies and appendages as well protection from the environment predation. This ubiquitous yet evolutionarily variable feature has been instrumental in facilitating adoption variety lifestyles exploitation ecological niches across environments. Throughout radiation produced more than one million described modern species, adaptability afforded by segmentation exoskeletons led to diversity is unrivalled amongst animals. However, because limited extensibility exoskeleton chitin cuticle components, they must be periodically shed replaced with new larger ones, notably accommodate growing individuals encased within. In the face of rising global demand and unsustainable production methods, cultivated crustacean meat (CCM) is proposed as an alternative means to produce delicious lobster, shrimp, crab products. Cultivated requires starting stem cells that may vary in terms potency propensity proliferate or differentiate into myogenic (muscle-related) tissues. Recognizing regenerating limbs are a non-lethal source tissue harbor relevant cells, we selected those crayfish Cherax quadricarinatus our model. To investigate cell activity, conducted RNA-Seq analysis across six stages claw regeneration (four pre-molt two post-molt stages), along with histology real-time quantitative PCR (qPCR). Our results showed while genes related energy production, muscle hypertrophy, exoskeletal cuticle synthesis dominated stages, growth factor receptors (FGFR, EGFR, TGFR, BMPR) proliferation (Cyclins, CDKs, Wnts, C-Myc, Klf4, Sox2, PCNA, p53) were upregulated before molt. Pre-molt upregulation several occurred peaks; Stages 2 4. We therefore propose limb tissues, particularly larger Stage 4, present prolific for CCM development.

Cytochrome P450s (CYP450s) are a versatile superfamily of enzymes known to undergo rapid evolution. Conserved CYP450s crustaceans those associated with ecdysteroidogenesis: synthesising breaking down the active moult hormone, 20-hydroxyecdysone. Four Shed show overlapping expression metamorphosis adult tissues, suggesting pleotropic functions multiple within ornatus.

Eriocheir sinensis demonstrates representative discontinuous growth during molting, while muscle is most obvious tissue exhibiting this property. Methods In work, a comprehensive analysis gene expressions E. between post-molt inter-molt stages were performed by integrating ATAC-seq RNA-seq techniques.
