ABSTRACT Aluminum oxide nanoparticles (Al 2 O 3 NPs) are widely utilized in vaccine manufacturing and other medical preparations. Melatonin has numerous effects as an antioxidant anti‐apoptotic. The purpose of this study was to examine the beneficial impact melatonin on Al NPs toxicity spinal cord. Forty male rats were divided into four groups: Group I, negative controls (received standard diet distilled water); II, 30 mg/kg bw NPs); III, + 10 melatonin); IV, melatonin). All treatments administered daily for 28 days by gastric gavage. After that, all sacrificed, then, samples from different cords subjected histopathological, biochemical, immunohistochemical analyses. markedly elevated malondialdehyde 8‐hydroxydeoxyguanosine while inhibiting superoxide dismutase catalase. also induced histological alterations both gray white matter manifested neuronal degeneration, vacuolation, axonal ballooning, fusion myelin sheaths. Furthermore, results displayed a strong positive expression caspase‐3. Conversely, significantly mitigated increasing activities enzymes 8‐hydroxydeoxyguanosine. Moreover, alleviated most reduced caspase‐3 immunoreactivity. Collectively, could protect cord mitigate NPs‐induced neurotoxicity.
