Atherosclerosis is a chronic inflammatory disease that causes degenerative and productive changes in the arteries. The resulting atherosclerotic plaques restrict vessel lumen, causing blood flow disturbances. Plaques are formed mainly large- medium-sized arteries, usually at bends forks where there turbulence flow. Depending on their location, they can lead to various states such as myocardial infarction, stroke, renal failure, peripheral vascular diseases, or sudden cardiac death. In this work, we reviewed literature early detection of atherosclerosis markers application photodynamic therapy atherosclerosis-related diseases. Herein, described roles C-reactive protein, insulin, osteopontin, osteoprotegerin, copeptin, TGF-β cytokine family, amino acid homocysteine. Also, discuss role microelements iron, copper, zinc, Vitamin D promoting formation plaque. Dysregulation administered compounds associated with an increased risk atherosclerosis. Additionally, taking into account pathophysiology plaque formation, believe maintaining homeostasis range biomarkers mentioned article crucial for slowing down process development stability already formed.

Cardiovascular diseases represent the principal cause of death and comorbidity among people with diabetes. Ferroptosis, an iron-dependent non-apoptotic regulated cellular characterized by lipid peroxidation, is involved in pathogenesis diabetic cardiovascular diseases. The susceptibility to ferroptosis hearts possibly related myocardial iron accumulation, abnormal metabolism excess oxidative stress under hyperglycemia conditions. Accumulating evidence suggests can be therapeutic target for This review summarizes ferroptosis-related mechanisms novel choices targeting pathways. Further study on ferroptosis-mediated cardiac injury enhance our understanding pathophysiology provide more potential choices.

Derived from hazelnuts, hazel leaf has been utilized in traditional folk medicine for centuries countries such as Portugal, Sweden, and Iran. In our previous investigations, we conducted a preliminary assessment of the polyphenol extract (referred to ZP) identified nine compounds, kaempferol chlorogenic acid, its composition. ZP shown promising properties an antioxidant anti-inflammatory agent. Our research revealed that protective effects against cisplatin-induced acute kidney injury (AKI). We comprehensive examination both pathological ultrastructural aspects found effectively ameliorated renal tissue lesions mitigated mitochondrial damage. Moreover, significantly suppressed malondialdehyde levels while increasing glutathione catalase concentrations kidneys AKI-induced mice. decreased number apoptotic cells pro-apoptotic protein expression mice human tubular epithelial (HK-2). Furthermore, treatment with increased proteins marking anti-ferroptosis, GPX4, FTH1, FSP1, experiments vivo vitro. elucidated underlying mechanisms ZP’s actions, revealing inhibitory effect on Yap phosphorylation regulation Lats expression, which exert influence kidneys. inhibiting Hippo pathway compromised nephroprotective vitro studies. summary, this shows exhibits renoprotective properties, reducing oxidative damage, apoptosis, ferroptosis by targeting pathway.

Obesity has emerged as a major public health challenge in the 21st century, contributing to rising prevalence of metabolic syndrome (MetS), cluster interrelated risk factors. These factors include obesity or abdominal obesity, type 2 diabetes mellitus (T2DM), hypertension (HTN), and dyslipidaemia. In this review, we will explore important aspects regulation dynamics lipoprotein metabolism see how they underlie each these risks. The identification of ferroptosis represents a pivotal advancement in the field cell death research, revealing an entirely novel mechanism cellular demise and offering new insights into initiation, progression, therapeutic management various diseases. Ferroptosis is predominantly induced by intracellular iron accumulation, lipid peroxidation, or impairments antioxidant defense system, culminating membrane rupture consequent death. Studies have associated with wide range diseases, enhancing our comprehension its underlying mechanisms, we can formulate innovative strategies, thereby providing renewed hope for patients.

Obesity is increasingly prevalent worldwide and linked to metabolic diseases, such as insulin resistance (IR) type 2 diabetes mellitus (T2DM), due excessive free fatty acids (FFAs). Although lifestyle changes are effective, they often prove be insufficient initial treatments for obesity. Additionally, while surgical pharmacological interventions available, not entirely safe or effective. Recently, interest has grown in utilizing food waste plant-derived phenolic compounds their health benefits, presenting a promising avenue managing obesity its related disorders. Indeed, many studies have examined the potential inhibitory effects of natural extract on adipocyte differentiation lipid accumulation. This study focused evaluation standardized extracts obtained from red oranges olive leaf 3T3-L1 murine pre-adipocyte functionality. Red orange (ROE) (OLE), alone combination, were tested assess anti-obesity anti-inflammatory effects, well therapeutic benefits. Three vitro models established investigate (I) differentiation; (II) mature hypertrophic adipocytes challenged with palmitic acid (PA) erastin (ER), respectively; (III) erastin-induced cytotoxicity pre-adipocytes.

Metabolic syndrome is increasing worldwide, particularly in rural communities, where residents have a higher risk of exposure to pesticides. We investigated whether six commonly used agricultural pesticides on corn and soy fields possess adipogenic metabolic disruption activity. Exposure two these pesticides, the herbicides acetochlor metolachlor, induced adipogenesis vitro mouse 3T3-L1 preadipocytes. The most potent compound, acetochlor, was selected for further studies zebrafish. Acetochlor morphological malformations lethality zebrafish larvae with an EC50 7.8 µM LC50 12 µM. at 10 nM resulted lipid accumulation when simultaneously fed high cholesterol diet. To decipher molecular mechanisms behind action, we preformed transcriptomic lipidomic analysis exposed animals. combined omics results suggested that increased Nrf2 activity response reactive oxygen species, as well peroxidation ferroptosis. discovered structurally shares chloroacetamide group known inhibitors glutathione peroxidase 4 (GPX4). Computational docking covalently binds active site GPX4. Consistent this prediction, Gpx efficiently repressed by zebrafish, whereas increased. propose disrupts homeostasis inhibiting activity, resulting peroxidation, 4-hydroxynonenal, which turn activate Nrf2. Because among other acetanilide herbicides, also contain group, inhibition may represent novel, common initiating event disruption.
