Purpose: Cervical cancer (CC) has the fourth highest incidence and mortality rate among female cancers. Lactate is a key regulator promoting tumor progression. Long non-coding RNAs (lncRNAs) are closely associated with cervical (CC). The study was aimed to develop prognostic risk model for based on lactate metabolism-associated lncRNAs determine their clinical value. Patients Methods: In this study, CESC transcriptome data were obtained from TCGA database. 262 genes extracted MsigDB (Molecular Characterization Database). Then, correlation analysis used identify LRLs. Univariate Cox regression performed afterwards, followed by least absolute shrinkage selection operator (LASSO) multiple analysis. 10 finally identified construct score model. They divided into two groups of high low according median scores. predictive performance models assessed Kaplan-Meier (K-M) analysis, subject work characteristics (ROC) univariate multivariate analyses. To assess utility model, we functional enrichment immune microenvironment mutation column line graph generation. Results: We constructed consisting LRLs at CC. observed that high-risk populations strongly poor survival outcomes. Risk an independent factor CC prognosis mutational load. Conclusion: developed metabolism it predict CC, which could guide facilitate progress new treatment strategies disease monitoring in patients. Keywords: cancer, metabolism, long RNA, bioinformatics,

Background: Cellular senescence is a novel hallmark of cancer associated with patient outcomes and tumor immunotherapy. However, the value cellular senescence-related long non-coding RNAs (lncRNAs) in predicting prognosis immunotherapy response for stomach adenocarcinoma (STAD) patients needs further investigation. Methods: The transcriptome corresponding clinical information STAD genes were, respectively, downloaded from Cancer Genome Atlas (TCGA) CellAge databases. Differential expression analysis coexpression were performed to obtain lncRNAs. Univariate regression least absolute shrinkage selection operator (LASSO) Cox conducted establish lncRNA prognostic signature (CSLPS). Next, survival curve, ROC nomogram developed assess capacity predictive models. Moreover, principal component (PCA), gene set enrichment (GSEA), microenvironment (TME), mutation burden (TMB), microsatellite instability (MSI), immune dysfunction exclusion (TIDE) score between high- low-risk groups. Results: A CSLPS involving fifteen lncRNAs (REPIN1-AS1, AL355574.1, AC104695.3, AL033527.2, AC083902.1, TYMSOS, LINC00460, AC005165.1, AL136115.1, AC007405.2, AL391152.1, SCAT1, AC129507.1, AL121748.1, ADAMTS9-AS1) was developed. According nomogram, risk model based on an independent factor could predict 1-, 3-, 5-year overall patients. GSEA suggested that high-risk group mainly Toll-like receptor, JAK/STAT, NOD-like chemokine signaling pathways. Further revealed better had higher TMB, proportion high (MSI-H), infiltration, lower TIDE scores. Conclusion: fifteen-CSlncRNA outcomes, may be more sensitive

Drugs, which treat various diseases, are essential for human health. However, developing new drugs is quite laborious, time-consuming, and expensive. Although investments into drug development have greatly increased over the years, number of approvals each year remain low. Drug repositioning deemed an effective means to accelerate procedures because it can discover novel effects existing drugs. Numerous computational methods been proposed in repositioning, some were designed as binary classifiers that predict drug-disease associations (DDAs). The negative sample selection was a common defect this method. In study, reliable scheme, named RNSS, presented, screen out pairs diseases with low probabilities being actual DDAs. This scheme considered information from k-neighbors one network, including their drug. Then, scoring system set up evaluate diseases. To test utility three classic classification algorithms (random forest, bayes network nearest neighbor algorithm) employed build using samples selected by RNSS. cross-validation results suggested such provided nearly perfect performance significantly superior those traditional previous schemes.
