Alzheimer's Disease (AD) is the most prevalent type of dementia and characterized by presence senile plaques neurofibrillary tangles. There are various theories concerning causes AD, but connection between viral bacterial infections their potential role in pathogenesis AD has become a fascinating area research for field. Various viruses such as Herpes simplex virus 1 (HSV-1), Epstein-Barr (EBV), Cytomegalovirus (CMV), influenza viruses, Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2), well bacteria Chlamydia pneumoniae (CP), Helicobacter pylori (HP), Porphyromonas gingivalis (P. gingivalis), Spirochetes eukaryotic unicellular parasites (e.g., Toxoplasma gondii), have been linked to due ability activate immune system, induce inflammation increase oxidative stress, thereby leading cognitive decline AD. In addition, microRNAs (miRNAs) might play crucial mechanisms these pathogens since they utilized target protein-coding genes, allowing evasion, maintaining latency, suppressing cellular signaling molecules. Also, can regulate gene expression human cells. This article provides an overview association infectious agents, with focus on which may be related These findings suggest important areas further explored future studies.

Abstract The posterior cingulate cortex (PCC) is a key hub of the default mode network underlying autobiographical memory retrieval, which falters early in progression Alzheimer’s disease (AD). We recently performed RNA sequencing post-mortem PCC tissue samples from 26 elderly Rush Religious Orders Study participants who came to autopsy with an ante-mortem diagnosis no cognitive impairment but collectively displayed range Braak I–IV neurofibrillary tangle stages. Notably, cognitively unimpaired subjects displaying high stages may represent resilience AD pathology. Transcriptomic data revealed elevated synaptic and ATP-related gene expression Stages III/IV compared I/II, suggesting these pathways be related resilience. also mined profiles for small non-coding micro-RNAs (miRNAs), regulate mRNA stability underexplored potential mechanism through fine-tuning within complex cellular networks. Twelve miRNAs were identified as differentially expressed between I/II III/IV. However, extent levels all associated subject demographics, neuropsychological test performance and/or neuropathological diagnostic criteria this cohort was not explored. Here, we report that total 667 are significantly (rho > 0.38, P < 0.05) variables. There significant positive correlations miRNA age, perceptual orientation speed. By contrast, higher correlated negatively semantic episodic memory. Higher 15 lower I–II 47 III–IV, additional mechanistic influences Pathway analysis showed enrichment operating lysine degradation fatty acid synthesis metabolism. Finally, demonstrated 12 resilience-related versus predicted mRNAs amyloid processing, tau inflammation. In summary, demonstrate dynamic state wherein differential intact elders. posit relationships inform transcriptional alterations relevant protective (resilience) or pathogenic (pre-clinical prodromal) responses pathogenesis thus therapeutic targets.

Mild cognitive impairment (MCI) is characterized by a decline in functioning without significant interference daily activities. Its high heterogeneity and elevated conversion rate to dementia pose challenges for accurate diagnosis monitoring, highlighting the urgent need identify methodologies focused on early detection intervention of MCI. Due their biological characteristics, microRNAs (miRNAs) are potential candidates as non-invasive molecular markers identification assessment MCI progression. Therefore, this study, we conducted meta-analysis miRNAs commonly deregulated MCI, focusing expression profiles plasma, serum, extracellular vesicle samples. Our analysis identified eight upregulated miRNAs, including hsa-miR-149-3p, four downregulated such Let-7f-5p. Notably, hsa-miR-149-3p emerged central node interaction networks, suggesting its crucial role regulating cellular processes relevant Additionally, pathway revealed enrichment associated with transcriptional regulation neurodegeneration. results underscore circulating open possibility new that enable more monitoring disease Validating Let-7f-5p, along identifying functional specific context essential establish relevance. This work contributes understanding miRNA profile mild using easily accessible samples, which could be useful development various strategies aimed at preventing or delaying individuals risk developing dementia, Alzheimer's disease.

Anti-amyloid therapies (AATs) are increasingly being recognized as promising treatment options for Alzheimer’s disease (AD). Amyloid-related imaging abnormalities (ARIAs), small areas of edema and microbleeds in the brain presenting abnormal signals MRIs patients with AD, most common side effects AATs. While ARIAs asymptomatic, they can be associated symptoms like nausea, headache, confusion, gait instability and, less commonly, more serious complications such seizures death. Cerebral amyloid angiopathy (CAA) has been found to a major risk ARIA development. The identification sensitive reliable non-invasive biomarkers CAA an area AD research over years, but approval AATs, this taken on new urgency. This comprehensive review highlights several potential biomarkers, Aβ40, Aβ40/42, phosphorylated-tau217, neurofilament light chain, glial fibrillary acidic protein, secreted phosphoprotein 1, placental growth factor, triggering receptor expressed myeloid cells 2, cluster differentiation 163, proteomics, microRNA. Identifying staging even before its consequences detected via neuroimaging critical allow clinicians judiciously select appropriate candidates stratify monitoring, properly manage therapeutic regimens those experiencing symptomatic ARIAs, optimize achieve best outcomes. Future studies test plasma human beings evaluate predictive values individual markers severity.
