Acinetobacter baumannii, an opportunistic bacterium prevalent in various environment, is a significant cause of nosocomial infections ICUs. As the causative agent pneumonia, septicemia, and meningitis, A. baumannii typically exhibits multidrug resistance associated with poor prognosis, thus led to challenge for researchers developing new treatment prevention methods. This study involved development novel multi-epitope mRNA vaccine validation silico approaches was conducted. We screened 11 immunodominant epitopes cytotoxic T cells, 5 helper 10 Linear B-cell based on promising candidate proteins omp33-36, ompA ompW, selection these three reverse vaccinology screening previous work by other researchers. All predicted demonstrated strong antigenicity, immunogenicity without posing any potential harm humans. Additionally, high conservancy required cover different strains. epitopes, as well adjuvants, were constructed into final vaccine, which further assessed calculating its physicochemical properties. Next, we docked protein immune receptors analyzed complexes dynamic simulations evaluate affinity receptors. At last, sequence translated sequence. The results indicated capability eliciting robust humoral cellular responses, making it protection against targeted pathogen.

In parallel to the uncontrolled use of antibiotics, emergence multidrug-resistant bacteria, like Acinetobacter baumannii , has posed a severe threat. A. predominates in nosocomial setting due its ability persist hospitals and survive antibiotic treatment, thereby eventually leading an increasing prevalence mortality infection. With spectra drug resistance incessant collapse newly discovered new therapeutic countermeasures have been high demand. Hence, recent research shown favouritism towards long-term solution designing vaccines. Therefore, being realistic alternative strategy combat this pathogen, anti- Baumannii vaccines continued unearthing various antigens with variable results over last decade. Again, other approaches, including pan-genomics, subtractive proteomics, reverse vaccination strategies, promise for identifying promiscuous core vaccine candidates that resulted chimeric constructs. addition, integration basic knowledge pathobiology drug-resistant bacteria also facilitated development effective multiantigen As opposed conventional trial-and-error approach, incorporating silico methods studies, particularly network analysis, manifested great novel from proteome. Acinetobacter baumannii is an emerging opportunistic pathogen. It exhibits multi-, extreme-, and pan-drug resistance against several classes of antibiotics. Capsular polysaccharide (CPS or K-antigen) one the major virulence factors which aids A. in evading host immune system. K-antigens exploit Wzx/Wzy-dependent pathway that involves 13 different proteins for its assembly transport onto outer membrane. A total 64 (out 237 K-locus(KL) types) known K-antigen sugar repeating structures are discussed here classified into seven groups based on their initial sugars, QuiNAc4NAc, GalNAc, GlcNAc, Gal, QuiNAc/FucNAc, FucNAc, GlcNAc along with Leg5Ac7Ac/Leg5Ac7R. Thus, corresponding initializing glycosyltransferases (ItrA1, ItrA2, ItrA3, ItrA4, ItrB1, ItrB3, ItrA3 ItrB2) exhibit serotype specificity. Acinetobacter baumannii (A. baumannii) is a Gram-negative opportunistic pathogen widely attached to the surface of medical instruments, making it one most common pathogens nosocomial infection, and often leading cross-infection co-infection. Due extensive antibiotic pan-resistance, A. infection facing fewer treatment options in clinic. Therefore, prevention have become tricky global problem. The requirement for research development new strategy urgent. Now, non-antibiotic strategies are urgently needed. This review describes on vaccines antibacterial adjuvants, discusses advantages disadvantages different candidate tested vitro vivo, especially subunit protein vaccines, shows efficacy adjuvant drugs monotherapy.
