Abstract Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) caused the persistent disease 2019 (COVID-19) pandemic, which has resulted in millions of deaths worldwide and brought an enormous public health global economic burden. The recurring wave infections been exacerbated by growing variants SARS-CoV-2. In this study, virological characteristics original SARS-CoV-2 strain its concern (VOCs; including Alpha, Beta, Delta) vitro, as well differential transcriptomic landscapes multiple organs (lung, right ventricle, blood, cerebral cortex, cerebellum) from infected rhesus macaques, were elucidated. a stronger innate immune response host cells, VOCs markedly increased levels subgenomic RNA s, such N, Orf9b, Orf6, Orf7ab, are known antagonists inhibitors antiviral factors. Intriguingly, Alpha variant induced larger alteration abundance tissues monkeys than Beta Delta did. Moreover, hyperinflammatory state active shown ventricles up-regulation inflammation- immune-related RNAs. Furthermore, peripheral blood may mediate signaling transmission among to coordinate molecular changes individuals. Collectively, these data provide insights into pathogenesis COVID-19 at early stage infection VOCs.

The ongoing 2019 coronavirus disease pandemic (COVID-19), caused by severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2) and its variants, is a global public health threat. Early diagnosis identification of SARS-CoV-2 variants plays critical role in COVID-19 prevention control. Currently, the most widely used technique to detect quantitative reverse transcription real-time PCR (RT-qPCR), which takes nearly 1 hour should be performed experienced personnel ensure accuracy results. Therefore, development nucleic acid detection kit with higher sensitivity, faster greater important.Here, we optimized system components reaction conditions our previous approach using RT-RAA Cas12b.We developed Cas12b-assisted one-pot platform (CDetection.v2) that allows rapid 30 minutes. This was able up 5,000 copies/ml without cross-reactivity other viruses. During the COVID-19 pandemic, different SARS-CoV-2 variants of concern (VOC) with specific characteristics have emerged and spread worldwide. At same time, clinicians routinely evaluate results certain blood tests upon patient admission as well during hospitalization to assess disease severity overall status. In present study, we searched for significant cell count biomarker differences among patients affected Alpha, Delta Omicron VOCs at admission. Data from 330 were retrieved regarding age, gender, VOC, (WBC, Neut%, Lymph%, Ig%, PLT), common biomarkers (D-dimers, urea, creatinine, SGOT, SGPT, CRP, IL-6, suPAR), ICU death. Statistical analyses performed using ANOVA, Kruskal-Wallis test, two-way Chi-square, T-test, Mann-Whitney test logistic regression was where appropriate SPSS v.28 STATA 14. Age VOC significantly associated hospitalization, whereas groups found WBC, PLT, D-dimers suPAR. Our showed that throughout current not only but also laboratory parameters are used patient's status subject changes.

Evolution effective vaccines against SARS-CoV-2 provided multiple options prevent infection, but treatment forms remains difficult manage.
