Abstract One of the fundamental problems current transplantation is organ shortage. To address this issue, Extended Criteria Donor grafts are utilized. However, requires better preservation strategies and reliable methods assessing graft function. The objective work was to evaluate changes in kidney metabolome intact organs subjected three methods: normothermic ex vivo perfusion (NEVKP), hypothermic machine (HMP) static cold storage (SCS). chemical biopsy used for minimally invasive sampling metabolites, followed by analysis on liquid chromatography coupled a high-resolution mass spectrometry (LC-HRMS) platform. results revealed metabolites altered ischemia oxidative stress donor’s kidneys, as well compounds characterizing occurring perfused under conditions storage. metabolomic differences were also recorded immediately after reperfusion few days surgery. It clearly shown that all associated with different biochemical alterations. pattern NEVKP indicated sustaining enables activation physiological anti-oxidative anti-inflammatory mechanisms tissue. some protective known exhibit Janus’sface, which might be an important finding future design treatment. A metabolomics profile HMP SCS kidneys confirmed despite fact hypothermia lowers energy demand cells, it restricts repair mechanisms. studies lie ground experiments toward assessment improved directly from utilizing non-destructive approach. The can serve versatile tool discovery investigations, but translated into diagnostic monitor specific biomarkers quality future.

Hepatocellular carcinoma is the most common form of primary liver cancer and poses a significant challenge to medical community because its high mortality rate. In recent years, ferroptosis, unique cell death, has garnered widespread attention. Ferroptosis, which characterized by iron-dependent lipid peroxidation mitochondrial alterations, closely associated with pathological processes various diseases, including hepatocellular carcinoma. Long non-coding RNAs (lncRNAs), are type functional RNA, play crucial regulatory roles in variety biological processes. this manuscript, we review lncRNAs key aspects summarize research progress on ferroptosis-related

The skin is the largest organ in human body and made up of various cells structures. Over time, will age, which not only influenced by internal factors, but also external environmental especially ultraviolet radiation. Aging causes immune system weakening elderly, makes them more susceptible to dermatosis, such as type 2 inflammatory mediated pruritus. response this condition marked senescent consistently releasing low amounts pro-inflammatory cytokines through a senescence-associated secretory phenotype (SASP). This continuous inflammation may accelerate aging establish connection between diseases. In addition, two chronic pigmentation disorders, vitiligo chloasma, are associated with aging. Aged escape accumulate tissues, forming microenvironment that promotes cancer. At same "photoaging" caused excessive exposure radiation an important cause manuscript describes possible links inflammation, cancer suggests some treatment options.

Kidney disease is marked by complex pathological mechanisms and significant therapeutic hurdles, resulting in high morbidity mortality rates globally. A deeper understanding of the fundamental processes involved can aid identifying novel targets improving treatment efficacy. Current comprehensive data analyses indicate involvement methyltransferase-like 3 (METTL3) its role RNA N6-methyladenosine methylation various renal pathologies, including acute kidney injury, fibrosis, chronic disease. However, there a paucity thorough reviews that clarify functional METTL3 evaluate importance enhancing outcomes. This review seeks to systematically examine roles, mechanisms, potential clinical applications diseases. The findings presented suggest implicated etiology exacerbation disorders, affecting their onset, progression, malignancy, responsiveness chemotherapeutic agents through regulation specific genetic pathways. In conclusion, this underscores detrimental correlation between diseases, highlighting promise targeting METTL3. Additionally, it offers critical insights for researchers concerning diagnosis, prognosis, strategies conditions.
