Over the past two decades, epigenetic regulation has become a rapidly growing and influential field in biology medicine. Widely used rodent anxiety assays like the elevated plus maze (EPM) and open field test (OFT) are conflated with rodents' natural preference for dark over light environments or protected spaces. The EPM OFT have been decades but often criticized by behavioral scientists. Years ago, two revised were designed to improve upon "classic" tests excluding possibility avoid escape aversion. 3-D radial arm (3DR) (3Doft) utilize continual motivational conflict better model anxiety; each consist of an space connected ambiguous paths toward uncertain escape. Despite their utility, not caught on. This could be because no study yet has directly compared classic in same animals. To remedy this, we contrasted behavior from a battery (EPM, OFT, 3DR, 3Doft sociability test) mice defined genetically isogenic strain, environmentally postnatal experience. One major motivation this work is inform future studies offering transparent look at individual outcomes on these assays, as there one-size-fits-all assess anxiety-like behavior. Findings suggest that may sufficiently characterize differences across groups, 3DR advantageous investigating more nuanced such those stemming environmental factors. Finally, exposure multiple significantly affected sociability, highlighting concerns designing interpreting batteries tests.

Abstract Treatment response and resistance in major depressive disorder (MDD) show a significant genetic component, but previous studies had limited power also due to MDD heterogeneity. This literature review focuses on the factors associated with treatment outcomes MDD, exploring their overlap those clinically relevant symptom dimensions. We searched PubMed for: (1) genome-wide association (GWASs) or whole exome sequencing (WESs) that investigated efficacy MDD; (2) examining between specific dimensions; (3) GWASs of identified 13 one WES reporting several loci, genes, gene sets involved expression, immune system regulation, synaptic transmission plasticity, neurogenesis differentiation. Nine dimensions were poor studied by (anxiety, neuroticism, anhedonia, cognitive functioning, melancholia, suicide attempt, psychosis, sleep, sociability). Four genes both these dimensions: CGREF1 (anxiety); MCHR1 (neuroticism); FTO NRXN3 (sleep). Other overlapping signals found when considering suggestively outcomes. Genetic showed convergence at level biological processes, despite no replication variant level. The interest may point shared mechanisms potential targets for new treatments tailored individual patient’s clinical profile.
