Piezo channels, a class of mechanosensitive ion have emerged as key players in sensory transduction. Piezo1 and Piezo2 been implicated various physiological processes, including touch sensation nociception. Their association with migraine their role pain involving trigeminal nerve gained significant research attention. Following PRISMA guidelines, we performed systematic review the literature, on channels pain. From PubMed, Cochrane Library, Scopus, deemed 20 studies published between 2014 2023 appropriate to be included this review. emerges player pathogenesis, contributing meningeal nociception generation. In syndromes, particularly Piezo2, conditions, from corneal dental orofacial Mechanistic insights provide potential therapeutic targets for other conditions nerve. This accentuates emerging significance trigeminal-associated pain, underscoring cross-associations that interconnect migraine, neurons, well suggesting promising avenues targeted interventions future directions.
