Background: Tanshinone IIA (Tan IIA) is a lipophilic active constituent derived from the rhizomes and roots of Salvia miltiorrhiza Bunge (Danshen), common Chinese medicinal herb. However, clinical applications Tan are limited due to its poor solubility in water. Methods: To overcome this limitation, we developed calcium alginate hydrogel (CA) as hydrophilic carrier for IIA, which significantly improved solubility. We also prepared nanoparticles with pH-responsive properties explore their potential controlled drug delivery. The physicochemical IIA/CA were evaluated, including size, stability, release profile. utilized RNA sequencing further investigate underlying anticancer mechanisms nanoparticles. Results: demonstrated enhanced exhibited potent activity vitro. Additionally, showed promising behavior, beneficial applications. Further investigation into molecular revealed that effects mediated through apoptosis, ferroptosis, autophagy pathways. Conclusions: This study confirms while presenting an innovative approach enhance poorly soluble compound. use CA-based could be valuable strategy improving therapeutic efficacy cancer treatment.

Epigenetic and metabolic reprogramming alterations are two important features of tumors, their reversible, spatial, temporal regulation is a distinctive hallmark carcinogenesis. Epigenetics, which focuses on gene regulatory mechanisms beyond the DNA sequence, new entry point for tumor therapy. Moreover, drives hepatocellular carcinoma (HCC) initiation progression, highlighting significance metabolism in this disease. Exploring inter-regulatory relationship between epigenetic modification has become one hot directions current research. As viral etiologies have given way to dysfunction-associated steatotic liver disease (MASLD)-induced HCC, it urgent that complex molecular pathways linking them hepatocarcinogenesis be explored. However, how aberrant crosstalk modifications affects MASLD-induced HCC lacks comprehensive understanding. A better understanding linkages necessary improve treatment strategies. For reason, review examines interwoven landscape carcinogenesis context focusing regulating development interactions while also updating advances modification-based therapeutic drugs HCC.

Hepatic fibrosis (HF), a global issue that develops gradually during the repeated repair process of chronic liver injury caused by various factors, can progress to irreversible cirrhosis and hepatocellular carcinoma. Currently, there are no approved chemical or biological drugs for treatment fibrosis. Modern medicine primarily focuses on addressing underlying causes HF is classified as an accumulative disease in Yellow Emperor's Internal Classic, which earliest most influential extant medical classic China. Thousands years experience have accumulated large number effective Chinese medicines. In recent years, studies anti-liver effects mainly focused curative extracts compounds. Studies shown prevention through demonstrate comprehensive pharmacological effect involves multiple pathways targets. Therefore, further in-depth research this field both promising necessary. However, it crucial expand clinical application studies, current TCM predominantly limited laboratory investigations. review, we summarized effects, targets related mechanisms 13 traditional medicines past five order provide information development drugs.

Non-alcoholic fatty liver disease (NAFLD) or metabolic dysfunction-associated steatotic (MASLD) and steatohepatitis (NASH) are chronic hepatic conditions leading to hepatocellular carcinoma (HCC) development. According the recent “multiple-parallel-hits hypothesis”, NASH could be caused by abnormal metabolism, accumulation of lipids, mitochondrial dysfunction, oxidative endoplasmic reticulum stresses is found in obese non-obese patients. Recent translational research studies have discovered new proteins signaling pathways that involved not only development NAFLD but also its progression NASH, cirrhosis, HCC. Nevertheless, mechanisms HCC developing from precancerous lesions yet been fully elucidated. Now, it particular importance start focusing on discovery novel molecular mediate alterations glucose lipid which leads steatosis. The role mTOR has recently attracted attention. goals this review (1) highlight genetic protein contributions NAFLD/NASH; (2) investigate how scientific findings might outline process causes NASH-associated HCC; (3) explore reliable biomarkers/targets NAFLD/NASH-associated hepatocarcinogenesis.
