Abstract Extrachromosomal circular DNA (eccDNA) is a new biomarker and regulator of diseases. However, the role eccDNAs in large‐artery atherosclerotic (LAA) stroke remains unclear. Through high‐throughput circle‐sequencing technique, length distribution, genomic characteristic motifs feature plasma eccDNA from healthy controls (CON) patients with LAA were analysed. Then, potential functions annotated investigated using GO KEGG pathway analyses. EccDNAs mapped to reference genome showed SHN3 BCL6 unique transcription factors. The genes differentially expressed between CON mainly involved axon/dendrite/neuron projection development maintenance cellular structure via Wnt, Rap1 MAPK pathways. Moreover, played regulation coagulation fibrinolysis, there five (Chr2:12724406‐12724784, Chr4:1867120‐186272046, Chr4:186271494‐186271696, Chr7:116560296‐116560685 Chr11:57611780‐5761192). Additionally, POLR2C AURKA carried by ecDNAs (eccDNA size >100 kb) significantly associated stroke. Our data firstly revealed characteristics genes, suggesting novel mechanism

Ischemic stroke (IS), primarily caused by cerebrovascular obstruction, results in severe neurological deficits and has emerged as a leading cause of death disability worldwide. Recently, there been increasing exploration the neuroprotective properties inert gas argon.
