Abstract Papillary thyroid carcinoma (PTC), the most common cancer, has been linked to various molecular alterations. This study focuses on microRNA‐223‐3p, whose upregulated expression in PTC tissues appears enhance tumor growth and cellular dysfunctions. Our findings demonstrate that microRNA‐223‐3p significantly promotes cell proliferation, invasion, migration induces epithelial‐mesenchymal transition (EMT). Additionally, neurofibromatosis type 2 (NF2) is identified as a direct target, suggesting could be crucial pathogenesis may offer target for therapeutic intervention.

The incidence of ulcerative colitis (UC) is increasing annually, and UC has a serious impact on patients’ lives. Polysaccharides have gained attention as potential drug candidates for treating in recent years. Huaier (Trametes robiniophila Murr) fungus that been used clinically more than 1000 years, its bioactive polysaccharide components reported to possess immunomodulatory effects, antitumour potential, renoprotective effects. In this study, we aimed examine the protective effects mechanisms (HP) against UC. Based H2O2-induced oxidative stress model HT-29 cells dextran sulphate sodium salt (DSS)-induced model, demonstrated polysaccharides significantly alleviated DSS-induced (weight loss, elevated disease activity index (DAI) scores, colonic shortening). addition, HP inhibited inflammation intestinal barrier damage. It also promoted expression mucin Muc2. Furthermore, reduced abundance harmful bacteria Escherichia-Shigella beneficial Muribaculaceae_unclassified, Anaerotruncus, Ruminococcaceae_unclassified regulate flora disturbance caused by DSS. Nontargeted metabolomics revealed intervention would modulate metabolism promoting levels 3-hydroxybutyric acid, phosphatidylcholine (PC), phosphatidylethanolamine (PE). These results had ability mitigate suppressing inflammation, maintaining barrier, modulating flora. findings will expand our knowledge how functions offer theoretical foundation using prebiotic prevent

Objective This study evaluated the clinical efficacy of Huaier granules combined with targeted therapy plus immunotherapy in patients unresectable hepatocellular carcinoma (HCC) who had not undergone systemic treatment. Methods Patients HCC and no prior treatments were recruited from Hospital Traditional Chinese Medicine Xinjiang First Affiliated Medical University between March 2022 July 2023. received (exposure group) or without Granules (non-exposure group). The primary endpoint was progression-free survival (PFS), secondary endpoints including 6-month PFS rate, Symptom Severity Quantitative Response, EORTC QLQ-HCC18 Score, safety. Results mPFS exposure group 8.9 months compared to 5 non-exposure (P = 0.001; HR 0.50). rates 66.7% 34.1% for groups, respectively 0.001). rate TCM symptom classification higher (87.50% vs 59.09%; P also showed improvement fatigue 0.023). Extrahepatic metastasis an independent prognostic factor (HR 1.77; 0.016), while reduced risk disease progression by 47% 0.53; 0.006). No significant differences observed adverse events. most common events hypertension, proteinuria, abnormal liver function, diarrhea. Conclusion significantly prolong improve reducing patients. Subgroup analysis more pronounced benefits vascular invasion alcohol consumption, extending beyond 1 year.

Highlights•Huaier can be an effective drug to reduce oxaliplatin-induced hepatotoxicity.•ROS-mediated oxidative stress is involved in hepatotoxicity.•Huaier relieves liver stress.•Inhibition of PI3K pathway reverses the hepatoprotective effect Huaier.AbstractHepatotoxicity caused by anticancer medication oxaliplatin (OXA) significantly restricts its clinical use and raises risk damage. Huaier, a fungus found China, has been demonstrated have various beneficial effects adjuvant therapy for cancer. However, preventive impact Huaier against OXA-induced hepatotoxicity still unknown. The potential molecular pathways behind activity were investigated current study Mice intraperitoneally injected with 10 mg/kg OXA once week six consecutive weeks establish injury model. (2 g/kg, 4 8 g/kg) was administered weekly mice gavage weeks. Commercial kits used determine contents glutathione, catalase, superoxide dismutase, malondialdehyde. Quantitative real-time polymerase chain reaction (qRT-PCR) Western blotting assess on expression pathway. exhibited good protective dose-dependent manner, which connected suppression stress, according results biochemical index detection histological staining analysis. In addition, could counteract PI3K/AKT signaling Moreover, activation eradicated LY294002. These findings imply that decreasing minimize injury, establishing groundwork lessen chemotherapy-induced practice.

Tumor xenograft animal models play a crucial role in hepatocellular carcinoma (HCC) research. Mice are time consuming, laborious and expensive while zebrafish tumor cost-effective effortless. However, the development of orthotopic for HCC embryos has been challenging due to small size livers. In this study, we utilized 7-day-old goldfish as hosts successfully established an model Through injecting fluorescence labeled cells into liver goldfish, could visualize proliferation migration vivo. addition, found that temperature 36 °C was better cell survival larvae compared 28 °C, assessed by EdU TUNEL assays. Moreover, macrophage infiltration be evaluated neutral red staining. Finally, efficacy targeted therapy drug Sorafenib traditional Chinese medicine, Huaier granules, alone or combination model. We showed best against terms infiltration, polarization well proliferation, metastasis apoptosis. conclusion, proposed opens new avenues related research, including evaluation progression, interactions immune microenvironment, efficacy, screening anti-tumor drugs.
