Chemotherapy-induced peripheral neuropathy (CIPN) is a dose-limiting painful that occurs commonly during cancer management, which often leads to the discontinuation of medication. Previous studies suggest α9α10 nicotinic acetylcholine receptor (nAChR)-specific antagonist αO-conotoxin GeXIVA[1,2] effective in CIPN models; however, related mechanisms remain unclear. Here, we analyzed preventive effect on neuropathic pain long-term oxaliplatin injection-induced model. At end treatment, lumbar (L4-L6) spinal cord was extracted, and RNA sequencing bioinformatic analysis were performed investigate potential genes pathways GeXIVA[1,2]. inhibited development mechanical allodynia induced by chronic treatment. Repeated injections for 3 weeks had no mice’s normal threshold or locomotor activity anxiety-like behavior, as evaluated open field test (OFT) elevated plus maze (EPM). Our results identified 209 differentially expressed (DEGs) model, simultaneously injecting with altered 53 DEGs. These reverted significantly enriched immune-related represented cytokine–cytokine interaction pathway. findings could be therapeutic compound oxaliplatin-induced management.

Palmitoylethanolamide (PEA) emerged over the years as a promising approach in management of chronic pain. Despite fact that efficacy micron-size PEA formulations appears to be time-dependent, optimal timing has not yet been elucidated. This systematic review and meta-analysis aim estimate possible advantage an extended treatment relief The literature search was conducted consulting scientific databases, identify clinical trials which administered for at least 60 days, pain assessed by Visual Analogue Scale (VAS) or Numeric Rating (NRS). Nine studies matched required criteria, total 742 patients involved. showed statistically clinically significant intensity reduction after days supplementation, compared 30 (1.36 points, p < 0.01). secondary analysis revealed weighted NRS/VAS score decrease 2.08 points within first month treatment. These two obtained scores corresponded 35.1% month, followed further 35.4% during second month. Overall, these results confirm relevant time-depended pain-relieving effect therefore treatment, especially patient with incomplete management.

Neuropathic pain, which refers to pain caused by a lesion or disease of the somatosensory system, represents wide variety peripheral central disorders. Treating neuropathic is quite demanding, primarily because its intricate underlying etiological mechanisms. The nervous system relies on microglia maintain balance, as they are associated with serving primary immune responses in brain next cell communication. Ferroptosis, driven phospholipid peroxidation and regulated iron, vital mechanism death regulation. Neuroinflammation can be triggered ferroptosis microglia, contributes release inflammatory cytokines. Conversely, neuroinflammation induce iron accumulation resulting microglial ferroptosis. Accumulating evidence suggests that neuroinflammation, characterized glial activation substances, significantly exacerbates development pain. By inhibiting ferroptosis, it may possible prevent subsequently alleviate homopentameric α7 subtype neuronal nicotinic acetylcholine receptor (α7nAChR) has potential suppress activation, transitioning M1 an M2 phenotype, facilitating anti-inflammatory factors, ultimately reducing Recent years have witnessed growing recognition regulatory role α7nAChR could target for treating This review summarizes mechanisms related progress according recent research. Such exploration will help elucidate relationship between α7nAChR, provide new insights into management.

Neuropathic pain causes tremendous biological and psychological suffering to patients worldwide. Environmental enrichment (EE) is a promising non-pharmacological strategy with high cost-effectiveness reduce neuropathic support rehabilitation therapy. Three researchers reviewed previous studies determine the efficacy of EE for research how improves through neuroinflammation. ALS is a progressive neurodegenerative disease that has serious impact on patients and their caregivers. For long time in the past, was considered painless largely ignored by clinicians. Describing complexity needs of pain symptoms from perspective can provide most intuitive direction for future research. The purpose our research to explore experience with amyotrophic lateral sclerosis (ALS), reference better understanding patients. From April 2023 May 2023, 27 experiencing Peking University Third Hospital who met diagnostic criteria "Chinese Guidelines Diagnosis Treatment sclerosis" were interviewed means objective sampling. content analysis method used describe changes since (amyotrophic sclerosis), factors aggravate pain, measures cope needs. interview results included 3 themes 11 subthemes. (1) Pain diverse: type when occurs, change intensity, pain; (2) Individualized coping measures: posture adjustment, medication, physical therapy, warmth, emotional regulation; (3) Patients lack pain: insufficient source knowledge, single orientation solution. nature, location aggravating China are complicated, which should be paid attention clinical staff scientific researchers. situation management not optimistic, vast majority been effectively alleviated. Neuropathic pain after spinal cord injury lacks any effective treatments, often leading to chronic pain. This study tested whether the daily administration of fully characterized polyphenolic extracts from grape stalks and coffee could prevent both reflexive non-reflexive neuropathic in cord-injured mice by modulating neuroimmune axis. Female CD1 underwent mild contusion received intraperitoneal weeks one, three, six post-surgery. Reflexive responses were assessed weekly for up 10 weeks, was evaluated at study's end. Neuroimmune crosstalk investigated, focusing on glial activation expression CCL2/CCR2 CX3CL1/CX3CR1 supraspinal pain-related areas, including periaqueductal gray, rostral ventromedial medulla, anterior cingulate cortex, amygdala. Repeated treatments prevented mechanical allodynia thermal hyperalgesia, also modulated Moreover, they reduced gliosis regulated signaling. Overall, combination polyphenols these may offer a promising pharmacological strategy modifying central sensitization markers, not only site but key regions implicated data highlight potential injury-induced
