Cannabidiol (CBD), a non-psychoactive cannabinoid, has attracted significant research interest due to its antioxidant, anti-inflammatory, and neuroprotective properties. As versatile scaffold in drug discovery, CBD been widely explored for developing novel therapeutics. In this study, we synthesized evaluated the anti-tyrosinase activity of CBD-based thiosemicarbazones. Structure–activity relationship (SAR) analyses were conducted assess impact various functional groups on tyrosinase inhibition, including an evaluation inhibitory kinetics selected compounds. The derivatives demonstrated potent with comparable kojic acid, standard inhibitor. Given crucial role melanin biosynthesis, these findings suggest that thiosemicarbazones could serve as promising candidates managing tyrosinase-related disorders, hyperpigmentation melanogenesis-related conditions. Background: Melasma is a chronic hyperpigmentary disorder exacerbated by sun exposure, characterized asymptomatic hyperpigmented macules and patches. Its pathogenesis involves ultraviolet B (UVB)-induced activation of melanogenesis pathways, including arachidonic acid (AA) metabolism, alpha-melanocyte-stimulating hormone (α-MSH) secretion, melanin synthesis. This study aimed to evaluate the efficacy multi-ingredient skin-lightening formulation (3% tranexamic acid, 2% ascorbyl glucoside, 2.5% arbutin, 5% niacinamide) compared 10% vitamin C preparation 4% hydroquinone (HQ) in suppressing UVB-induced AA, α-MSH, production keratinocyte-melanocyte co-culture model. Methods: Keratinocyte-melanocyte co-cultures were pretreated with formulation, C, or HQ before UVB irradiation (150 mJ/cm2). Distilled water served as negative control. Biomarkers (AA, melanin) quantified using enzyme-linked immunosorbent assay (ELISA) fluorometric assay.
