Abstract The seeds of oilseed rape ( Brassica napus L.) are used in large-scale production one the most health-promoting plant oils food industry, as well for animal feed and biofuel production. Thus, increasing yield this crop is crucial economic ecological importance. However, conventional breeding programs slow, laborious time-consuming. Hence, along with discovery possibility to apply CRISPR/Cas technology edit genomes accelerate process, much effort has been put into applying study specific genes biosynthetic pathways, especially species many gene copies such B. . Brassica napus (B. napus) is a globally significant oilseed crop, making substantial contribution to both human oil and livestock feed production. Enhancing seed weight crucial for improving rapeseed yield; however, only limited number of weight-related genes have been functionally validated in B. thus far. UBIQUITIN-SPECIFIC PROTEASE 15 (UBP15) belongs the ubiquitin protease pathway plays maternal role prolonging development Arabidopsis. The potential utilization UBP15 enhancing yield has remained unexplored until now. In this study, we identified orthologs investigated its functionality using CRISPR-Cas9 system. We generated mutant plants with multiple editing types targeting Bnaubp15s successfully isolated T-DNA-free homozygous lines that exhibited edits across four homologs BnaUBP15 T2 generation plants. Our preliminary data demonstrated mutation BnaUBP15s significantly reduced size, weight, plant height while noticeably increasing primary branches. These findings not provide evidence further elucidating molecular mechanism underlying regulation size by but also offer promising novel germplasm architecture.
