This study aimed to characterize the sperm kinematic values with high frames per second, define subpopulation structure of a horse and donkey compare them. A total 57 fresh semen ejaculates (26 Spanish 16 Arabian breeds 10 donkeys) were collected subsequently analyzed for parameters using Computer-aided motility analysis ISAS®v1.2 system Spermtrack® 10-μm depth counting chamber. Sequences recorded at 250 eight automatically evaluated. All showed significant differences between breeds. evaluated excellent characteristics, significantly higher all donkeys compared horses except beat-cross frequency. Donkey was faster linear than horse. Regarding differences, had average path velocity, curvilinear frequency rapid, but Arab more linear. The principal component three subpopulations in ejaculate different characteristic dominant both straight, beat (38.2 41.7%, respectively), whereas lowest slowest non-linear sperms. This, plus slight distribution these horses, found. In conclusion, permitted have new interpretation motile species breed differences. More so, future works on spermatozoa should take into account that may interfere alter real performed.

In donkeys, the use of frozen-thawed sperm for artificial insemination (AI) leads to low fertility rates. Furthermore, donkey produce a large amount reactive oxygen species (ROS), and post-AI inflammation induces formation neutrophil extracellular traps (NETosis), which further generates many more ROS. These high ROS levels may induce lipid peroxidation in plasma membrane, thus affecting its integrity. Enzymatic non-enzymatic antioxidants, mainly found seminal (SP), are responsible maintaining redox balance. However, this fluid is removed prior cryopreservation, thereby exposing cells oxidative stress. The exogenous addition antioxidants freezing medium can reduce detrimental effects caused by generation. Therefore, aim study was evaluate how different reduced glutathione (GSH) concentrations (control, 2 mM, 4 6 8 10 mM) fresh affect their cryotolerance. Total progressive motility, kinematic parameters motile subpopulations were significantly (p < 0.05) from control treatments containing mM GSH, but not at lower concentrations. Plasma acrosome membrane integrity, mitochondrial potential (MMP) intracellular superoxide (O2-) affected > any GSH concentration. Interestingly, however, or percentages viable with overall compared control. conclusion, able tolerate concentrations, differs what has been observed other species. This antioxidant capacity suggests that could be important during impact using like improve resilience freeze-thawing limited

Good sperm production is a notable requirement for subjects intended reproduction, particularly in endangered species, and it has been demonstrated that horse stallions, this correlated to testicular volume. The present study, which involved Martina Franca jacks, aimed determine whether, also breed, there correlation between the total number (TSN) Testes were measured with both ultrasound caliper. Testicular volume was calculated using two different formulas: one representing of an ellipsoid developed describe round-shaped testicles. average concentration 380.14 ± 254.58 × 106/mL, while TSN 16.34 7.76 109. Our findings evidenced significant (r > 0.75; p < 0.05) only formula V (cm3) = 33.57 H − 56.57 testes. Moreover, significance data obtained (VTs-us 315.03 25.83 cm3) but not In conclusion, can be suggestive good fertility jacks; thus, parameter could considered when selecting breeding animals.

The purpose of this study is to investigate the optimal framerate (FR) and use different counting chambers for improving CASA-Mot technology in Andrology. Images were captured at 500 fps, then segmented analyzed several ranges FRs (from 25 250) define asymptotic point that as an FR. This work was replicated using based capillarity (disposable) or drop displacement (reusable) their effects on motility results kinematic values samples under experimental conditions. α value (asymptote corresponding FRo) exponential curve 150.23 a VCL 130.58 mm/s, far from 98.89 mm/s 50 fps (the highest FR used by most current systems). Our have shown that, when reusable chambers, type depth influence. In addition, obtained depending area image inside chamber types.
