Abstract Receptor tyrosine kinases (RTKs), a category of transmembrane receptors, have gained significant clinical attention in oncology due to their central role cancer pathogenesis. Genetic alterations, including mutations, amplifications, and overexpression certain RTKs, are critical creating environments conducive tumor development. Following discovery, extensive research has revealed how RTK dysregulation contributes oncogenesis, with many subtypes showing dependency on aberrant signaling for proliferation, survival progression. These findings paved the way targeted therapies that aim inhibit crucial biological pathways cancer. As result, RTKs emerged as primary targets anticancer therapeutic Over past two decades, this led synthesis validation numerous small molecule kinase inhibitors (TKIs), now effectively utilized treating various types. In manuscript we provide comprehensive understanding context We explored alterations specific receptors across different malignancies, special dedicated examination current inhibitors, highlighting potential therapies. By integrating latest evidence, seek elucidate pivotal biology efficacy inhibition promising treatment outcomes.

Abstract: Bone homeostasis is a dynamic equilibrium state of bone formation and absorption, ensuring skeletal development repair. immunity encompasses all aspects the intersection between immune systems, including various signaling pathways, cytokines, crosstalk cells under both homeostatic pathological conditions. Therefore, as key cell types in immunity, macrophages can polarize into classical pro-inflammatory M1 alternative anti-inflammatory M2 influence body environment, participating regulation metabolism playing roles homeostasis. not only act precursors osteoclasts (OCs), differentiate mature OCs, but also secrete cytokines to promote resorption; while osteogenic factors, stimulating differentiation mineralization osteoblast mesenchymal stem (MSCs), subsequently increase formation. Once polarization imbalanced, resulting dysregulation will cause inflammatory stimulation, release large amount factors affecting metabolism, leading conditions such osteoporosis (OP), rheumatoid arthritis (RA), steroid-induced femoral head necrosis (SANFH). In this review, we introduce pathways related macrophage polarization, well their relationships with OB, OC, MSC. We discuss diseases imbalance, regulating them, which may help develop new methods for treating metabolic disorders. Keywords: macrophage,

Colorectal cancer (CRC) is one of the main fatal cancers. Cell signaling such as Janus kinase/signal transducer and activator transcription (JAK/STAT) substantially influences process gene expression cell growth. Long non-coding RNAs (lncRNAs) play regulatory roles in signaling, proliferation, fate. Hence, lncRNAs can be considered biomarkers The inhibitory or activating effects different on JAK/STAT pathway regulate proliferation tumor suppression. Additionally, immune responses which a role immunotherapy. Mechanisms CRC via regulation mainly include invasion, metastasis, apoptosis, adhesion, control inflammation. More profound findings are warranted to specifically target terms activation suppression hindering proliferation. Here, understand lncRNA cross-talk through pathway, we collected related vitro vivo data. Future insights may pave way for development novel diagnostic tools, therapeutic interventions, personalized treatment strategies patients.
