: Non-alcoholic fatty liver disease (NAFLD) is one of the most common diseases worldwide. Reactive oxygen species (ROS), as potent oxidants in cells, have been shown to promote development NAFLD. Previous studies reported that for ROS-induced cellular oxidative stress, promoting lipid droplet (LD) accumulation associated with antioxidation process. However, regulatory role LDs relieving stress poorly understood. Here, we showed Perilipin 5 (PLIN5), a key LD protein related mitochondria–LD contact, reduced ROS levels and improved mitochondrial function HepG2 cells. Both mRNA PLIN5 were significantly increased cells hydrogen peroxide or lipopolysaccharide (LPS) treatment (p < 0.05). Additionally, overexpression promoted formation up-regulated function-related genes such COX CS. Knockdown PLIN5, meanwhile, opposite effects. Mitochondria are peculiar organelles whose proper function depends on the crosstalk between two genomes, mitochondrial and nuclear. The human genome (mtDNA) encodes only 13 proteins; nevertheless, its expression is essential for cellular homeostasis, as mtDNA-encoded proteins constituents of respiratory complexes. In addition, mtDNA results in production RNA molecules, which influence cell physiology once released from mitochondria into cytoplasm. As a result, dysfunctions may lead to pathologies humans.
