Neoplastic disorders, particularly malignant carcinomas, are complex systemic diseases characterized by unregulated cellular proliferation, the invasion of adjacent tissues, and potential metastasis to distant bodily sites. Among diverse spectrum cancer subtypes, melanoma is a highly aggressive form cutaneous originating in melanocytes, pigment-producing cells resident skin. This malignancy distinguished its rapid uncontrolled growth, as well propensity for vital organs, thereby posing significant challenges therapeutic intervention prognostication. Early detection crucial optimizing patient outcomes, diagnosis at an advanced stage often yields poor prognosis limited treatment options. Diagnostic modalities encompass comprehensive clinical evaluations dermatologists; radiological imaging techniques such ultrasonography, magnetic resonance (MRI), computed tomography (CT) scans; excisional biopsies accurate histopathological assessment. Malignant typically treated with surgery remove tumor, followed immunotherapy enhance immune response, targeted therapy tumors specific genetic mutations, chemotherapy stages, radiation manage metastasis, other adjunct therapies. review presents properties possible effects against quercetin found literature explores, based on observed physicochemical biological activity, development topical formulation application. Quercetin naturally occurring flavonoid compound abundant various plant-based food sources, including apples, onions, berries, citrus fruits, has exhibited promising antiproliferative, antioxidant, anticancer properties. Its distinctive biochemical structure enables effectively neutralize reactive oxygen species modulate key carcinogenic pathways, rendering it candidate managing tumors, melanoma.

Both cutaneous melanoma (CM) and uveal (UM) represent important causes of morbidity mortality. In this review, we evaluate the available knowledge on differences similarities between melanoma, focusing epidemiological aspects risk factors. Uveal is a rare condition but most prevalent primary intra-ocular malignant tumor in adults. Cutaneous other hand, significantly more common. While frequency has increased last decades worldwide, incidence remained stable. Although both tumors arise from melanocytes, they are very distinct entities biologically, with complex varied etiologies. conditions encountered frequently by individuals fair phenotype. ultraviolet-radiation an important, well-documented factor for development CM, shown not to be specific UM. ocular melanomas seem inherited independently, there reported cases concomitant same patient.

Melanoma is a malignant, highly metastatic neoplasm showing increasing morbidity and mortality. Tumor invasion angiogenesis are based on remodeling of the extracellular matrix (ECM). Selective inhibition functional components cell-ECM interaction, such as hyaluronic acid (HA), metalloproteinases (MMPs), integrins, may inhibit tumor progression enhance efficacy combination treatment with immune checkpoint inhibitors (ICIs), chemotherapy, or immunotherapy. In this review, we combine results different approaches targeting elements in melanoma preclinical clinical studies. The identified limitations many approaches, including side effects, low selectivity, toxicity, indicate need for further studies to optimize therapy. Nevertheless, significant progress expanding our understanding biology development targeted therapies holds great promise early developed several decades ago metastasis through ECM targeting.

Aceclofenac (ACF) is a widely used non-steroidal anti-inflammatory drug (NSAID) known for its effectiveness in treating pain and inflammation. Recent studies have demonstrated that ACF possesses antiproliferative properties, inhibiting the growth of cancer cells various cell lines. Citronellol, monoterpenoid alcohol found essential oils, exhibits antioxidant properties activities such as acetylcholinesterase inhibition. In this study, objective was to formulate evaluate an aceclofenac/citronellol oil nanoemulsion effects on melanoma. The optimal concentrations citronellol oil, Tween 80, Transcutol HP were determined using pseudoternary phase diagram. formulated nanoemulsions characterized droplet size, zeta potential, thermophysical stability, vitro release. selected formula (F1) consisted (1 gm%), 80 (4 gm%). F1 exhibited spherical appearance with high content, small acceptable negative potential. amorphous state confirmed by Differential Scanning Calorimetry, while FTIR analysis indicated homogenous solubility. showed significant activity, lower IC50 value compared aceclofenac or alone. Flow cytometric revealed cycle arrest increased apoptosis induced nanoemulsion. silico provided insights into molecular mechanism underlying observed antitumor activity. conclusion, developed potent cytotoxicity pro-apoptotic effects, suggesting potential repurposed agent melanoma treatment. future plan, further animal model research validation suggested.

Fucoidan, a sulfate polysaccharide obtained from brown seaweed, has various bioactive properties, including anti-inflammatory, anti-cancer, anti-viral, anti-oxidant, anti-coagulant, anti-thrombotic, anti-angiogenic, and anti-Helicobacter pylori properties. However, the effects of low-molecular-weight fucoidan (LMW-F) on melanoma cell lines three dimensional (3D) culture models are not well understood. This study aimed to investigate LMW-F A375 human cells cryopreserved biospecimens derived patients with advanced melanoma. Ultrasonic wave was used fragment Fucus vesiculosus into smaller LMW-F. MTT live/dead assays showed that inhibited proliferation in both patient-derived explants 3D-printed collagen scaffold. The PTEN/AKT pathway found be involved anti-melanoma fucoidan. Western blot analysis revealed reduced phosphorylation Bcl-2 at Thr 56, which associated prevention anti-apoptotic activity cancer cells. Our findings suggested could enhance chemotherapy improve outcomes resistance.
