Abstract Background This study aimed to explore the expression, molecular mechanism and its biological function of potassium two pore domain channel subfamily K member 1 (KCNK1) in bladder cancer (BC). Methods We integrated large numbers external samples (n = 1486) assess KCNK1 mRNA expression levels collected in-house 245) for immunohistochemistry (IHC) experiments validate at protein level. Single-cell RNA sequencing (scRNA-seq) analysis was performed further cellular communication. The transcriptional regulatory mechanisms were explored by ChIP-seq, ATAC-seq ChIA-PET data. Highly expressed co-expressed genes (HECEGs) used potential signalling pathways. Furthermore, immunoassay, clinical significance docking calculated. Results significantly overexpressed BC (SMD 0.58, 95% CI [0.05; 1.11]), validated level (p < 0.0001). Upregulated exhibited highly distinguishing ability between control (AUC 0.82 [0.78–0.85]). Further, scRNA-seq revealed that predominantly clustered epithelial cells tended increase with differentiation. involved communication mainly through MK pathway. Secondly, transcription start site (TSS) showed promoter-enhancer interactions three-dimensional space, while being transcriptionally regulated GRHL2 FOXA1. Most HECEGs enriched cell cycle–related metabolism–related pathways membrane activity. associated infiltration various immune cells. Immunotherapy chemotherapy (docetaxel, paclitaxel vinblastine) more effective patients high group. correlated age, pathology grade pathologic_M patients. Conclusions BC. Bladder carcinoma (BC) is the most prevalent malignancy of urinary system. These cancers are primarily seen in adults > 60 years old and mostly present with microscopic or frank hematuria obstruction However, these rare can be found hernias. This report discusses a rare, localized bladder urothelial (UC) manifestation. The patient had presented lower abdominal pain several times. no accurate diagnosis was made due to unspecified features. Evidence regarding the association between tea consumption and bladder cancer (BC) risk is inconsistent. This study aimed to increase our knowledge of by using international data from Bladder Cancer Epidemiology Nutritional Determinants Consortium. The estimated multilevel multivariable logistic regression analysis adjusted for multiple (non-)dietary factors. Association BC observed (odds ratio, OR = 0.72, 95% confidence interval, CI 0.65-0.80) compared non-tea drinkers. Stratified analyses based gender smoking status yielded similar results. showed no significant black green across models, while herbal linked a reduced (OR 0.59, 0.36-0.96). Background: Proteins with different functions, such as Hepatocyte growth factor activator inhibitor type 1 (HAI-1), Stathmin (STMN-1), and Tenascin C (TN-C), whose activity has been observed in various types of cancers, inspired our study bladder cancer (BC) patients. The aim the was to evaluate selected parameters their combinations diagnosis BC. took into account degree invasiveness malignancy Based on analysis Receiver Operating Characteristic Curve (ROC), diagnostic value single potential indicators BC assessed. Patients Methods: research material consisted urine samples from patients BC, a control group without urological diseases. concentrations examined were measured using an immunoenzymatic method. Results: Statistically significant higher HAI-1 (p ≤ 0.001), STMN-1 (≤0.001) TN-C (0.002) found compared group. Strong relationships shown between these parameters. ROC analyses showed that best parameter for detecting is STMN-1, combination HAI-1+STMN-1. highest obtained HAI-1+STMN-1 high (sensitivity 82%, specificity 91%). Conclusions: Preliminary studies have utility markers especially However, learn more about contribution progression cancer, it would be appropriate continue studies.
