Background: Alchornea laxiflora (Benth.) Pax & K. Hoffm. (A. laxiflora) is utilized as a traditional herb for treating several diseases. Objective: Our study aims to identify the active phytochemical candidates from A. and analyses predict their anticancer activity mechanism by employing network pharmacology, molecular docking, dynamics (MD). Methods: The phytoconstituents of were retrieved literature, phytoconstituent-related targets implicated in hepatocellular carcinoma (HCC) collected respective databases. Computational methods employed recognize essential compounds, hub gene targets, signaling pathways. Results: has identified 12 potentially bioactive 150 potential anti-HCC 15 laxiflora. Molecular docking results recognized better binding energy values below −5.6 kcal/mol. Further, MD simulations three top-scoring protein–ligand complexes (MAPK—3-acetylursolic acid, AKT1—quercetin, AKT1—3-acetylursolic acid) allowed us validate results, evaluate stability complexes, associated conformational changes. Abstract Hepatocellular carcinoma (HCC), as the most prevalent liver malignancy, is annually diagnosed in more than half a million people worldwide. Hepatocellular carcinoma (HCC), the most prevalent form of liver cancer, is third leading cause mortality globally. Patients with HCC have a poor prognosis due to fact that emergence symptoms typically occurs at late stage disease. In addition, conventional biomarkers perform suboptimally when identifying in its early stages, heightening need for identification new and more effective biomarkers. Using metabolomics lipidomics approaches, this study aims identify serum patients cirrhosis (LC). Serum samples from 20 cases LC were analyzed using ultra-high-performance liquid chromatography-Q Exactive mass spectrometry (UHPLC-Q-Exactive-MS). Metabolites lipids are significantly altered between identified. These include organic acids, amino TCA cycle intermediates, fatty bile glycerophospholipids, sphingolipids, glycerolipids. The significant variability was observed concentrations glycerophospholipids. context cases, there notable increase levels phosphatidylethanolamine triglycerides, but acids phosphatidylcholine exhibited substantial decrease. it all identified metabolites superior area under receiver operating characteristic (ROC) curve comparison alpha-fetoprotein (AFP). pathway analysis these revealed acid, lipid, energy metabolism as impacted pathways. Hepatocellular carcinoma (HCC) is one of the leading causes cancer-related mortality worldwide. The AKT pathway often activated in HCC cases, and a longer exposure to tyrosine kinase inhibitors such as sorafenib may lead over-activation pathway, resistance. Here, we studied efficacy new generation allosteric inhibitor, vevorisertib, alone or combination with sorafenib. To identify specific adverse effects related background cirrhosis, used diethylnitrosamine (DEN)-induced cirrhotic rat model. Vevorisertib was tested vitro on Hep3B, HepG2, HuH7 PLC/PRF cell lines. Rats were treated weekly intra-peritoneal injections DEN for 14 weeks obtain cirrhosis fully developed HCC.
