Background: Gliomas are the most malignant tumors of central nervous system. One factors in their high drug resistance is avoiding programmed death (PCD) induction. This related to overexpression intracellular survival pathways: PI3K-Akt/PKB-mTOR and Ras-Raf-MEK-ERK. Apoptosis autophagy co-existing processes due interactions between Bcl-2 beclin-1 proteins. Their complex may be a molecular “toggle-switch” PCD types. The aim this research was investigate role Bcl-2:beclin-1 glioma cell elimination through combined action LY294002 sorafenib. Methods: Drug cytotoxicity estimated with an MTT test. type evaluated using variant microscopy techniques (fluorochrome staining, immunocytochemistry, transmission electron microscopy), as well formation protein localization. Molecular analysis indicators conducted immunoblotting, immunoprecipitation, ELISA testing. SiRNA used block expression. Results: results showed inhibitors simultaneous application resulted apoptosis becoming dominant. accompanied by changes location tested Conclusions: “Switching” PI3K Raf opens new therapeutic perspectives against gliomas.

Abstract Background Glioblastoma (GBM) is the most common malignant primary brain tumor. Emerging reports have suggested that racial and socioeconomic disparities influence outcomes of patients with GBM. No studies to date investigated these controlling for isocitrate dehydrogenase (IDH) mutation O‐6‐methylguanine‐DNA methyltransferase (MGMT) status. Methods Adult GBM were retrospectively reviewed at a single institution from 2008 2019. Univariable multivariable complete survival analyses performed. A Cox proportional hazards model was used assess effect race status priori selected variables known relevance survival. Results In total, 995 met inclusion criteria. Of these, 117 (11.7%) African American (AA).
