The aim of this study was to evaluate the impact diabetes mellitus type 2 (DM2) on male endocrine system Zucker Diabetic Fatty (ZDF) rats. Sexually mature ZDF rats were divided a lean (control) and obese group, had confirmed by blood tests. For in vivo experiment, fasting collected obtain plasma. In case vitro experiments, testicular fragments cultured for 24 h, culture medium collected. concentrations testosterone (T), androstenedione (A4), dehydroepiandrosterone (DHEA-S), estradiol (E2), follicle-stimulating hormone (FSH) luteinizing (LH) quantified plasma ELISA method, while cholesterol (CHOL) assessed spectrophotometrically. A significant decline T (36.31 %), A4 (25.11 %) FSH (26.99 as well increase CHOL E2 (36.17 observed comparison control. Under conditions, decrease (23.35 accompanied an group compared CHOL, LH, T, DHEA no differences observed. Our results suggest that except all steroid biomolecules synthetized normally tissue, however dramatic disturbance at level. We may conclude DM2 has negative effects systemic secretion these alterations are more pronounced combination with obesity.

BACKGROUND: Gonadotropin preparations, particularly human chorionic gonadotropin (hCG), are commonly used to induce ovulation and treat reproductive disorders in women, albeit with associated side effects. Low-molecular-weight allosteric agonists of the luteinizing hormone receptor (LHR), such as thieno[2,3-d]pyrimidine derivatives, offer a potential alternative. AIM: This study aims compare effects TP03 hCG on ovarian weight, corpus luteum formation, plasma levels estradiol, progesterone, immature female rats pre-treated Follimag®. It also examines their impact gene expression related LHR steroidogenesis. MATERIALS AND METHODS: were administered 48 h after Follimag® injection at dose 20 mg/kg (i.p.) 15 IU/rat (s.c.), respectively. Parameters assessed 1, 2, 4, 8, 16, 24 administration. Plasma measured via ELISA, was analyzed using real-time PCR. RESULTS: increased progesterone blood, steroidogenic genes encoding cholesterol-transporting protein StAR cytochromes CYP11A1 CYP17A1. stimulated formation (16–24 treatment). The temporal dynamics its stimulating similar those hCG, although magnitude slightly inferior gonadotropin. TP03-induced decrease blood estradiol aromatase ovaries more moderate. Unlike which suppressed 8 treatment, maintained high expression, preserving sensitivity endogenous hormone. CONCLUSIONS: exhibits an inducer milder steroidogenesis than reduces risks developing hyperstimulation syndrome resistance gonadotropins.

Luteinizing hormone (LH) and human chorionic gonadotropin (CG), like follicle-stimulating hormone, are the most important regulators of reproductive system. They exert their effect on cell through LH/CG receptor (LHCGR), which belongs to family G protein-coupled receptors. Binding induces interaction LHCGR with various types heterotrimeric proteins (Gs, Gq/11, Gi) β-arrestins, leads stimulation (Gs) or inhibition (Gi) cyclic adenosine monophosphate-dependent cascades, activation phospholipase pathway (Gq/11), also formation signalosomes that mediate mitogen-activated protein kinases (β-arrestins). The efficiency selectivity intracellular cascades by different gonadotropins varies, is due differences in ligand-binding site LHCGR. Gonadotropin signaling largely depends status N- O-glycosylation LH CG, homo- heterodimeric complexes, cell-specific microenvironment presence autoantibodies it, allosteric mechanisms implementation these influences, multiplicity sites loci development low-molecular-weight profiles pharmacological activity, can be used medicine for correction disorders assisted technologies, promising. These other issues regarding hormonal regulation summarized discussed this review.

The “cafeteria diet,” which includes an excess of saturated fats and easily digestible carbohydrates, leads to obesity is a risk factor for the development type 2 diabetes. Metformin (MF) often used correct diet-induced (DIO), but in some patients it causes serious side effects, requires reduction its doses, including through combined use with drugs that potentiate effects MF. A candidate role such intranasally administered insulin (INI), itself has restorative potential treatment metabolic disorders. goal work was study effectiveness three-week MF (100 mg/kg/day, perorally) INI two doses (1.5 6.0 IU/kg/day) correction hormonal disorders male rats DIO induced by diet”. It shown DIO, combination normalized body weight abdominal fat, restored glucose homeostasis, lipid metabolism, basal glucose-stimulated levels leptin. Compared monotherapy, more effectively sensitivity insulin, assessed decrease resistance index, also tolerance, value AUC0-120, integrated area under concentration curves tolerance test. at dose 1.5 IU/kg/day status thyroid system, disturbed while 6 worsened hypothyroid state, mainly due hyperactivation thyroid-stimulating hormone secretion gland it. Thus, parameters restoration functions relatively low IVI, does not have negative effect on axis, promising.

The search for natural biologically active substances that have a neuroprotective effect on cerebral ischemia-reperfusion is one of the urgent problems modern neuroscience and medicine. Intranasally administered insulin (IAI) has pronounced restorative various neurodegenerative diseases, but mechanisms its action therapeutic effects in ischemia not been studied well, including type 2 diabetes mellitus (DM2), which increases risk cerebrovascular dysfunction. aim work was to study IAI metabolic parameters inflammatory factors male rats with DM2 subjected two-vessel prolonged forebrain reperfusion, comparison non-diabetic animals. A long-term high-fat diet an injection low dose streptozotocin (25 mg/kg) used induce DM2, model global induced by occlusion both common carotids reperfusion (IR) 7 days ischemia. Two hours after end ischemia, were treated at 0.5 or 2.0 IU/rat, drug same doses daily subsequent days. It found prevents body weight loss nondiabetic diabetic underwent IR, also total cholesterol level proportion epididymal fat without IR. In explored reduces postprandial glucose content blood, indicates improvement tolerance, levels blood – C-reactive protein (at IU/rat/day) tumor necrosis factor-α (in IU/rat/day), reveals anti-inflammatory potential. Thus, course treatment induction followed leads weakens reactions may be demand correction ischemic stroke patients DM2.

Several reports, including our previous studies, indicate that hyperglycemia and diabetes mellitus exert differential effects on vascular function in males females. This study examines sex differences the of type 2 (T2D) an established monogenic model obesity-induced T2D, Zucker Diabetic Fatty (ZDF) rats. Acetylcholine (ACh) responses were assessed phenylephrine pre-contracted rings before after apocynin, a NADPH oxidase (NOX) inhibitor. The mRNA expressions aortic endothelial NOS (eNOS), key NOX isoforms also measured. We demonstrated following: (1) had contrasting vasorelaxation ZDF rats, impairing relaxation to ACh females while enhancing it male rats; (2) inhibition NOX, major source superoxide vasculature, restored female (3) eNOS NOX4 elevated (but not male) rat aortas compared their respective leans. highlights sexual dimorphism ACh-mediated aorta suggesting may play role impaired observed
