The aim of the preliminary study was to improve success cryopreservation stallion spermatozoa. effect different equilibration lengths and two techniques for freezing semen evaluated, based on motility viability parameters. Fresh diluted with BotuCrio extender divided into 3 groups equilibrated 0.5h, 2h, 4h. Subsequently, half straws each group were cryopreserved in vapours liquid nitrogen or automatic freezer. Results spermatozoa progressive showed decreasing trend increased time equilibration. Moreover, a significant difference (p < 0.05) percentage producing ROS found between frozen compared by freezer same time. Nevertheless, results this are character. Experiments using higher number individuals need be tested order find best procedure Slovak national horse breeds.

For decades now, sperm cryopreservation has been a pillar of assisted reproduction in animals as well humans. Nevertheless, the success varies across species, seasons, and latitudes even within same individual. With dawn progressive analytical techniques field genomics, proteomics, metabolomics, new options for more accurate semen quality assessment have become available. This review summarizes currently available information on specific molecular characteristics spermatozoa that could predict their cryotolerance before freezing process. Understanding changes biology result exposure to low temperatures may contribute development implementation appropriate measures assure high post-thaw quality. Furthermore, an early prediction or cryosensitivity lead establishment customized protocols interconnecting adequate processing procedures, techniques, cryosupplements are most feasible individual needs ejaculate.

The present review aims to provide an overview of the assay methods for quantification ROS and principal enzymatic antioxidants as biomarkers oxidative stress in erythrocytes spermatozoa small domestic ruminants. A complete literature search was carried out PubMed, Scopus World Wide Web using relevant keywords focusing on last five years (2018-2023). Among spectrophotometry, fluorometry chemiluminescence, most widely used method is fluorometry, probably because it allows simultaneously several ROS, different probes, with greater economic advantages. Regarding intracellular antioxidant enzymes, recent reports only spectrophotometric methods, many which use commercial kits. a less sensitive but cheapest suitable both samples are highly concentrated ruminant species. All considered this have been found be appropriate; general, differences related their costs sensitivity. The objective of this study was to evaluate the effect a tannin-rich extract from commercial Acacia mearnsii (MTE_0), and gallocatechin, flavonoid compound derived mearnsii, on long-term viability motility cryopreserved ovine semen. Six fresh ejaculates obtained six adult merino rams twice per week for three weeks were allocated five aliquots (0, 12.5, 25, 50, 100 µM gallocatechin added into Optidyl™ extender) before cooling cryopreservation. Effects MTE_0 post-thawed characteristics analyzed using computer-assisted semen analysis (CASA), (LIVE/DEAD® kit, Molecular Invitrogen, Waltham, MA), oxidative stress (2,7-dichlorodihydrofluorescein diacetate (H2DCFDA, Thermo Fisher®, MA)) reactive oxygen species (ROS), mitochondrial membrane potential (JC-1 MitoTracker, acrosomal integrity (lectin PNA), capacitation merocyanine 540 (M540) YO-PRO-1 dyes in flow cytometry. Data one-way ANOVA (IBM SPSS 21.0 Windows, Armonk, NY). Gallocatechin at 25 positively affected (p ≤ .001) kinematic parameters including average path velocity (VAP), progressive (VSL), beat cross frequency (BCF) Similarly, µM- improved sperm (live 21.99 ± 2.06%), reduced ROS levels (26.45 1.10%), mitigated premature (viable stable 20.08 1.48%) compared other treatments. addition resulted significant positive with extract. It is concluded that inclusion significantly reduces deterioration following This first introduce as an efficient antioxidant additive improve its quality during storage. Our findings will help post-thaw longevity. Future studies elucidate mechanism anti-oxidative action derivatives longevity are recommended.

The optimization of preservation protocols (refrigeration and freezing) in ovine species is necessary for a wider diffusion artificial insemination this species. Besides the ram sperm quality assays, characterization novel proteins could be crucial improving these employing biomarkers. protein transmembrane 95 (TMEM95) membrane associated with oocyte-sperm fusion previously described bull or mouse. However, has not yet been characterized until now. In work, different experimental groups based on functionality: capacitated, refrigerated at times (5 °C 24 h, 5 48 72 h), frozen-thawed samples were analyzed compared to initial (15 3 h) characterize expression its relationship other markers (motility, kinetic parameters, viability, apoptosis-like events, mitochondrial function, acrosome-reacted, zinc content as marker capacitation). addition, capacitation status was tested by Fluozin-3, fluorescent probe measuring used first time sperm. After induction, expected, acrosome reactive spermatozoa signature 2 significantly increased, while linearity (P < 0.05) decreased non-capacitated samples. Concerning TMEM95, profile increased after process, confirming status. Attending processes, semen progressively during liquid storage, significant decrease observed h according fast progressive motility linearity. TMEM95 showed same tendency, showing reduction respect control Finally, cryopreservation thawed suffered detrimental effect sample, concerting all studied parameters accomplished When we correlation markers, highest positive correlations low capacitated samples, such changes acrosome-reaction. Artificial insemination (AI) is used frequently in the breeding of sport horses, apart from Thoroughbreds. Most AIs are carried out with cooled semen rather than frozen because difficulties identifying a protocol that suitable for freezing most ejaculates and necessity to inseminate close ovulation short life thawed spermatozoa. More widespread use would improve biosecurity, allow greater choice stallions, offer more flexibility when managing deliveries stud. It even decrease amount antibiotics extenders, since volume smaller inseminated. However, there considerable variability cryosurvival spermatozoa different leading classification stallions as good or bad freezers. Improvements could be made at level stallion nutrition, collection regimen, extender, removal seminal plasma, cooling protocol, among others. Stallion sperm membranes highly susceptible lipid peroxidation, but research on antioxidants has failed identify an additive benefit all stallions. In future, biomarkers freezability aid cryopreservation.
