Zemplin Rabbit and Liptov Bold-Spotted are Slovak national breeds. Our aim was to characterize the sperm quality of these two breeds, as characteristics important for artificial insemination cryopreservation biodiversity conservation tools. For this evaluation, samples sexually mature (ZR) males (n = 6) Rabbits (LR) 4) were used. According progressive motility (PM) data (CASA), divided into groups: A (>30% PM) B (<30% PM). In addition PM (ZR-A: 48.6±3.8%, ZR-B: 16±3.2%, LR-A: 35.38±2.6%, LR-B: 4±2.2%), total (TM) 69.1±4.1%, 35.5±4.1%, 59.1±3%, 26.9±4.1%) morphological abnormalities 29.7±1.2%, 40±4%, 34.3±4.3%, 48.3±4.7%) also assessed using CASA system. The proportion dead/live, apoptotic oxidative damaged spermatozoa (Spz) by flow cytometry fluorescent dyes: SYBR-14, Sytox Green, Yo-Pro-1 CellROX respectively. results correspond values morphometry. Sperm with less than 30% does not show proper values, while higher is suitable further analysis use.

Cryopreservation plays a critical role in animal breeding and the conservation of endangered species, but it often compromises sperm characteristics such as morphology, motility, viability due to oxidative stress. This study explores antioxidative effect Mito-Tempo (MT) Berberine (BER) enhance post-thaw quality rabbits. Pooled rabbit samples were supplemented with different concentrations (0.0, 0.5, 5, 10, 50 µmol/L) MT BER. Sperm motility was evaluated using computer-assisted semen analysis, while viability, apoptosis, reactive oxygen species (ROS) levels, acrosome integrity, mitochondrial function assessed through flow cytometry. The results revealed that at 5 10 µmol/L BER significantly improved total progressive activity, compared control group. Furthermore, enhanced both effectively reduced ROS levels apoptosis. is first demonstrate protective effects on during cryopreservation. Cryopreservation reduces the antioxidant activity of spermatozoa and makes them more sensitive to damage caused by reactive oxygen species (ROS). The addition antioxidants freezing medium could prevent cryo-damage mitigating harmful effects ROS and, thus, protecting spermatozoa. This study aimed evaluate effect ellagic acid (EA) on rabbit sperm traits after freezing-thawing. Semen samples collected from New Zealand White males were cryopreserved in a BotuCrio (Nidacon, Sweden) supplemented with different concentrations (EA at 0, 0.5, 1.5 2.5 mM) using manual slow procedure. After thawing, motility parameters evaluated CASA. viability (DRAQ7), apoptosis (Yo-Pro-1), acrosome integrity (peanut agglutinin; PNA), intracellular (CellROX) mitochondrial (MitoTracker) flow cytometry. EA added all led significant reduction (P < 0.05) frozen-thawed cells. However, this was not reflected parameters. mM also yielded lower proportion apoptotic cells compared control group. In conclusion, supplementation semen extender demonstrated its antioxidative properties against oxidative during cryopreservation. Nevertheless, draw definitive conclusion regarding functionality, additional research is necessary.

The present study aimed to evaluate the effects of melatonin (MLT), glutathione (GSH), and their combination on ram semen quality after thawing. During eight weekly sessions, from three Merino rams was pooled, diluted with an egg-yolk-based extender, divided into four groups: control, 1 mM MLT, 5 GSH, MLT + GSH. Diluted evaluated before freezing process. supplementation GSH at had a deleterious effect total motility progressive (TPM), curvilinear velocity (VCL), straight-line (VSL), average-path (VAP), linearity (LIN), straightness (STR) increased slow spermatozoa (%). also negative TPM, VSL, STR in semen. In thawed semen, TPM compared control group. VSL lower group than Additionally, both antioxidants attenuated BCF. These results indicate that impairs or does not improve sperm kinetic parameters either They suggest combined plays protective role against these effects.

This research investigates the high prevalence of morphological defects in sperm a particular rabbit breed, namely Zemplin (ZR). We tried to uncover possible causes and consequences observed abnormalities. Ejaculate samples from ten adult males were divided into two qualitative groups according motility parameters revealed by CASA (computer-assisted analysis). Group A contained with total (TM) above 50% progressive (PM) 30%. B TM PM below these values. Viability (SYBR-14/DRAQ5TM SYTOXTM Green/DRAQ5TM staining) apoptosis (YO-PRO TM-1/DRAQ5 stainig) analyses through flow cytometry served as an additional insight characteristics groups. In both cases, we noted significant differences (P < 0.05) between which corresponded Both showed increased incidence abnormal spermatozoa, did not observe any them. Simultaneously, assessed practical implications on artificial insemination (AI), also, notice parameters. The use transmission electron microscopy enhanced our understanding at ultrastructural level. summary, study provides semen, highlighting variations quality While exist, they may necessarily translate AI outcomes. Our shows higher values cytoplasmic droplets ZR but it also that conditions do mean fundamental obstacle fertility case AI.
