Mulberry-derived postbiotics (MDP) have been scientifically proven to possess beneficial properties for human health, making them potential nutraceuticals. This study aims enhance the antioxidant and anti-inflammatory effect of MDP via submerged fermentation with Bacillus subtilis H4 (H4) amyloliquefaciens LFB112 (LFB112) bacterial strains. Our results demonstrated compelling evidence that combination two strains optimized time (24 hours), inoculum size (8%), substrate (10%) significantly increased 2,2-diphenyl-1-picrylhydrazyl (DPPH) scavenging activities. When compared non-fermented mulberry (NFM), above-optimized conditions showed substantially higher activities such as DPPH, 2,2'-Azino-bis (3-ethylebenzothiazoline-6-sulfonic acid) (ABTS), hydroxyl activities, reducing power capabilities. Moreover, a significant decrease in lipopolysaccharides (LPS)-induced reactive oxygen species (ROS) generated RAW264.7 cells. The antibacterial assays minimum inhibitory concentrations (MICs) bactericidal (MBCs) possesses activity against various pathogenic lowest MIC recorded 32.5 mg/ml S. pullorum CVCC519, while highest observed was 40 P. aeruginosa. MBCs were 55 aureus ATCC43300 CVCC519 respectively. Acute lung injury (ALI) is a syndrome caused by an excessive inflammatory response characterized intractable hypoxemia both inside and outside the lung, for which effective therapeutic drugs are lacking. Atractylodis rhizoma, traditional Chinese medicine, has excellent anti-inflammatory antiviral properties in addition to protecting integrity of cellular barrier. However, few studies rhizoma treatment ALI have been published, its mechanism action remains unclear. In present study, chemical composition ethanolic extract (EEAR) was initially clarified high performance liquid chromatography (HPLC), after it studied vivo using lipopolysaccharide (LPS)-induced rat model. Treatment with EEAR significantly reduced wet/dry (W/D) ratio, neutrophil infiltration, malondialdehyde (MDA) myeloperoxidase (MPO) formation, enhanced superoxide dismutase (SOD) glutathione (GSH) depletion rats ALI, thereby improving barrier function effectively reducing injury. addition, histopathological changes, decreased expression factors (such as tumor necrosis factor-α (TNF-α), interleukin-1 beta (IL-1β), inducible nitric oxide synthase (INOS), cyclooxygenase-2 (COX-2)), inhibited activation NF-κB signaling pathway, thus inflammation. found also reduce oxidative stress upregulating nuclear factor erythroid 2-related 2 (Nrf2) downstream proteins heme oxygenase-1 (HO-1) NADPH quinone acceptor oxidoreductase 1 (NQO-1). LPS-induced THP-1 cells vitro inhibition damage from promoting Nrf2 targets HO-1 NQO-1, molecular consistent observations. Therefore, we conclude that attenuates responses via TLR4/NF-κB Keap1/Nrf2 pathways alleviate suggesting potential drug candidate ALI.

Abstract The coronavirus disease 2019 (COVID-19) pandemic has impacted human lifestyles around the world, causing huge distress in terms of public health systems, emergency response capacity and economic development. causative agent COVID-19, severe acute respiratory syndrome 2 (SARS-CoV-2), is associated with involvement, cardiovascular-related diseases, ultimately causes multiple organ failure death severely affected individuals. Thus, effective prevention or early treatment COVID-19 critical. An vaccine offers a way out for governments, scientific community people worldwide, but we still lack drug therapies, including treatments COVID-19. This had led to high global demand many complementary alternative medicines (CAMs). Moreover, healthcare providers are now requesting information about CAMs that prevent, relieve, treat symptoms even alleviate vaccine-related side effects. Inflammatory bowel disease remains a significant challenge in clinical settings. This study investigated the therapeutic potential of sea cucumber ovum hydrolysates (SCH) dextran sulfate sodium (DSS)-induced colitis mouse model. SCH, defined by its elevated stability and solubility, with molecular weight below 1000 Da, significantly alleviated DSS-induced colitis, as evidenced enhanced splenic index, reduced colonic damage, diminished serum pro-inflammatory cytokines. Furthermore, macrogenomic analysis demonstrated that SCH increased beneficial gut microbes decreased bacteria. metabolomic tissues identified levels anti-inflammatory metabolites, such Phenyllactate, 2-Hydroxyglutarate, L-Aspartic acid, mice after oral administration SCH. In conclusion, represents promising candidate for treatment colitis.
