Abstract The molecular events underlying the development, manifestation, and course of schizophrenia, bipolar disorder, major depressive disorder span from embryonic life to advanced age. However, little is known about early dynamics gene expression in these disorders due their relatively late manifestation. To address this, we conducted a secondary analysis post-mortem prefrontal cortex datasets using bioinformatics machine learning techniques identify differentially expressed modules associated with aging diseases, determine time-perturbation points, assess enrichment quantitative trait loci (eQTL) genes. Our findings revealed early, mid, deregulation functional involved neurodevelopment, plasticity, homeostasis, immune response. This supports hypothesis that multiple hits throughout contribute disease manifestation rather than single early-life event. Moreover, time-perturbed were genetic affecting expression, highlighting role factors development phenotypes. emphasize importance investigating time-dependent perturbations before age onset elucidating mechanisms psychiatric disorders.

Although the negative association of tobacco smoking with osteoporosis is well-documented, little known regarding shared genetic basis underlying these conditions. In this study, we aim to investigate a architecture between and heel estimated bone mineral density (eBMD), reliable proxy for osteoporosis. We conducted comprehensive genome-wide cross-trait analysis identify correlation, pleiotropic loci causal relationship eBMD, leveraging summary statistics hitherto largest studies in European ancestry initiation (Nsmoker = 1 175 108, Nnonsmoker 493 921), heaviness (cigarettes per day, N 618 489), cessation (Ncurrent smoker 304 244, Nformer 843 028), eBMD (N 426 824). A significant global correlation was found (${r}_g$ -0.051, P 0.01), while failed or eBMD. Partitioning whole genome into independent blocks, observed 6 local signals 22q13.1 showing strongest regional correlation. Such overlap further supported by 71 identified meta-analysis. Mendelian randomization no effect (beta -0.003 g/cm2, 95% CI -0.033 0.027) -0.017 -0.072 0.038) on but putative predisposition being current associated lower compared former smokers -0.100 -0.181 -0.018). Our study demonstrates pronounced biological pleiotropy as well link status providing novel insights primary prevention modifiable intervention advocating individuals avoid, reduce quit early possible.
