Anoikis is a specialized form of programmed cell death and also related mitophagy process. We aimed to identify an anoikis mitophagy-related genes (AMRGs) prognostic model explore the role <i>SPHK1</i> in colon cancer (CC). Bioinformatic methods were used screen AMRGs. Based on these genes, all samples divided into different subtypes. Furthermore, LASSO was conducted optimize optimal risk score established evaluated. Finally, effects downregulated <i>SPHK1 </i>on CC proliferation, migration, invasion, investigated. AMRGs, subtype 1 2. An AMRGs signature containing three key (<i>SPHK1, CDC25C, </i>and <i>VPS37A</i>) that exhibiting predicting ability survival confirmed. Subtype2 low-risk groups exhibited better higher immune infiltration. Moreover, lower invasion ability, as well line (<i>P</i> &#60; 0.01). The exhibits promising patients with CC. might inhibit growth, through stimulating anoikis.

ABSTRACT Anoikis resistance in hepatocellular carcinoma (HCC) cells boosts survival and metastasis. This study aimed to establish an anoikis‐related genes (ARGs)‐based model for predicting HCC patients’ outcomes investigate the clinicopathological significance function of crucial ARGs. The transcriptional expression patterns cohorts were compiled from TCGA, GEO ICGC. Univariate LASSO multivariate analyses performed screen prognostic Gain‐ loss‐of‐function studies, RNA sequencing, mass spectrometry employed elucidate underlying mechanisms ARGs HCC. We established a five‐gene risk prognosis, with AUC value 0.812 1‐year survival. Among five genes, Rac family small GTPase 3 (RAC3) was upregulated relative adjacent normal tissues negatively correlated overall disease‐free patients Silence RAC3 resulted increased cell apoptosis diminished proliferation invasion. Mechanistically, we uncovered that binding SOX6 propelled advancement through NNMT‐mediated stimulation cAMP/MAPK/Rap1 signaling. In particular, EHop‐016, molecule inhibitor targeting RAC3, significantly suppressed progression.

Background Ovarian cancer (OC) is a highly lethal and aggressive gynecologic cancer, with an overall survival rate that has shown little improvement over the decades. Robust models are urgently needed to distinguish high-risk cases predict reliable treatment options for OC. Although anoikis-related genes (ARGs) have been reported contribute tumor growth metastasis, their prognostic value in OC remains unknown. The purpose of this study was construct ARG pair (ARGP)-based signature patients elucidate potential mechanism underlying involvement ARGs progression. Methods RNA-sequencing clinical information data were obtained from Center Genome Atlas (TCGA) Gene Expression Omnibus (GEO) databases. A novel algorithm based on pairwise comparison utilized select ARGPs, followed by Least Absolute Shrinkage Selection Operator Cox analysis signature. predictive ability model validated using external dataset, receiver operating characteristic curve, stratification analysis. immune microenvironment proportion cells analyzed high- low-risk seven algorithms. set enrichment weighted gene co-expression network performed investigate mechanisms occurrence prognosis. Abstract Considering the high fatality of hepatocellular carcinoma (HCC), current prognostic systems are insufficient to accurately forecast HCC patients' outcomes. In our study, nine anoikis‑related genes (PTRH2, ITGAV, ANXA5, BIRC5, BDNF, BSG, DAP3, SKP2, and EGF) were determined establish a risk scoring model using LASSO regression, which could be validated in ICGC dataset. Kaplan–Meier curves time-dependent receiver operating characteristic (ROC) curve analysis confirmed score possessed an accurate predictive value for prognosis patients. The high-risk group showed higher infiltration aDCs, macrophages, T-follicular helper cells, Th2 cells. Besides, PD-L1 was significantly compared low-risk group. Abstract: Hepatocellular carcinoma (HCC) stands as the prevailing form of primary liver cancer, characterized by a poor prognosis and high mortality rate. A pivotal factor in HCC tumorigenesis is epigenetics, specifically regulation gene expression through methylation. This process relies significantly on action proteins that modify methylation, including methyltransferases, their associated binding proteins, demethylases. These are crucial regulators, orchestrating methylation regulating enzymes corresponding proteins. orchestration facilitates reading, binding, detection, catalysis sites.
