Background/Objectives: BuZhong Yiqi Formula (BZYQF) has significant ameliorative effects on type 2 diabetes mellitus (T2DM). However, its efficacy in alleviating the hyposalivation caused by T2DM needs to be confirmed, and mechanism is unclear. Methods: Network pharmacology molecular docking were combined analyze which BZYQF alleviates T2DM-caused hyposalivation. A rat model was induced evaluate of BZYQF. The total saliva before after acid stimulation collected determine salivary flow rate alpha-amylase (sAA) activity. parotid (PG) submandibular glands (SMG) experimental rats removed perform histopathology observation, biochemical indicator determination, expression detection signaling molecules secretion pathway. Results: present study screened out 1014 potential targets regarding treatment T2DM. These mainly involved formation receptor complex, exercising neurotransmitter activity regulating secretion. They significantly enriched pathway β1-AR/PKA/AMY1 CHRM3/IP3R/AQP5. Furthermore, BZYQF, nine validated compounds able dock into active site β1-AR, three CHRM3. Further mechanistic studies revealed glucose lipid accumulation, acetylcholine content, improves pathological lesions inflammation, molecules, including PKA, IP3R, AQP5, CHRM3, AMY1 PG SMG 0.05). Conclusions: demonstrated alleviate improving metabolism activating This might provide a novel rationale strategy for diabetes-induced clinical setting.

Abstract Objectives This cross-sectional study aimed to investigate the oral health-related quality of life (OHRQoL), its associated factors, and prevalence possible sarcopenia in Thai well-maintained patients with end-stage renal disease (ESRD) undergoing peritoneal dialysis (PD). Materials Methods Data were collected from 63 participants PD at Banphaeo-Charoenkrung Hemodialysis Center. Dry mouth was evaluated through unstimulated salivary flow rate measurement self-reported xerostomia questionnaires. OHRQoL assessed using version Oral Health Impact Profile (OHIP-14). Statistical Analysis analyses conducted IBM SPSS Statistics 21.0. Descriptive statistics summarized participant characteristics, normality tested Kolmogorov–Smirnov test. Continuous variables expressed as medians interquartile ranges, while categorical presented frequencies percentages. The Mann–Whitney U test Fisher's exact used assess differences between groups. Partial Spearman's rank correlation examined variable relationships, logistic regression identified factors linked a higher negative impact on OHRQoL, adjusting for age, sex, body mass index, chair stand test, rate. A p-value < 0.05 considered significant. Results median age 59 years (range 27–79), 52.4%. OHIP-14 scores ranged 0 32, 4 13 lower (n = 31) 32) respectively. Those exhibited significantly proportion (p 0.01), longer 5-time 0.04) compared individuals OHRQoL. Correlation time handgrip strength (r –0.439, p 0.001) sex –0.351, 0.006) revealed. Multivariate showed significant association (odds ratio 0.018; 95% confidence interval: 0.001, 0.545; 0.02). There is an increasing trend towards the use of bioactive compounds in oral healthcare. Some have been identified as sialagogues, such mastic resin and its main isolate, α-pinene. These act acetylcholinesterase inhibitors (AChEIs), consequently salivary flow. With up to half global population experiencing dry mouth symptoms, there a clear need for new interventions. An vitro screening 148-compound library was conducted identify novel AChEIs. Following this, four were chosen vivo validation administered through nasal spray pumps olfactory stimulus healthy individuals (n = 12) evaluate their sialagogic effects. Saliva quantity, rheology, composition (total protein concentration MUC5B concentration) analysed before after compounds. This study confirmed α-pinene, basil, eugenol guaiacol AChEIs vitro. Elevated oxidative stress can play a pivotal role in autoimmune diseases by exacerbating inflammatory responses and tissue damage. In Sjögren’s disease (SjD), the contribution of pathogenesis remains unclear. To address this question, we created mice with tamoxifen-inducible conditional knockout (KO) critical antioxidant enzyme, superoxide dismutase 2 (Sod2), salivary glands (i-sg-Sod2 KO mice). Following tamoxifen treatment, Sod2 deletion occurred primarily ductal epithelium, showed significant downregulation expression. At twelve weeks post-treatment, from i-sg-Sod2 exhibited increased 3-Nitrotyrosine staining. Bulk RNA-seq revealed alterations gene expression pathways related to ribosome biogenesis, mitochondrial function, phosphorylation. Significant changes were noted genes characteristic gland ionocytes. The developed reversible glandular hypofunction. However, functional loss was not accompanied lymphocytic foci or circulating anti-nuclear antibodies. These data demonstrate that although localized cells insufficient for SjD development, it induced dysfunction. mouse resembles patients classified as non-Sjögren’s sicca will be valuable model deciphering oxidative-stress-mediated dysfunction recovery mechanisms.
