Neurological disorders present a wide range of symptoms and challenges in diagnosis treatment. Cannabis sativa, with its diverse chemical composition, offers potential therapeutic benefits due to anticonvulsive, analgesic, anti-inflammatory, neuroprotective properties. Beyond cannabinoids, cannabis contains terpenes polyphenols, which synergistically enhance pharmacological effects. Various administration routes, including vaporization, oral ingestion, sublingual, rectal, provide flexibility treatment delivery. This review shows the efficacy managing neurological such as epilepsy, neurodegenerative diseases, neurodevelopmental disorders, psychiatric painful pathologies. Drawing from surveys, patient studies, clinical trials, it highlights alleviating symptoms, slowing disease progression, improving overall quality life for patients. Understanding mechanisms can open up possibilities using this plant individual needs.

The endocannabinoid system (ECS) is one of the most important systems modulating functions in body. ECS, via cannabinoid (CB: CB1 and CB2) receptors, endocannabinoids occurring brain (e.g., anandamide (AEA) 2-arachidonoylglycerol (2-AG)) enzymes degrading (fatty-acid amide hydrolase (FAAH) monoacylglycerol lipase (MAGL)), plays a key role regulation mood anxiety. However, effects compounds on anxiety-related responses are complex yield mixed results depending type pharmacological manipulation (direct or indirect) as well kinds cannabinoids, dosage procedure. aim this study was to determine compare influence direct (via CB receptors ligands) indirect inhibition brain) modulation ECS function anxiety-like mice elevated plus maze (EPM) test. For purpose, first step experiments, we used selected ligands CB1, CB1/CB2 CB2 assess which types involved mice. Next, inhibitors FAAH (which breaks down AEA) MAGL 2-AG) more responsible for behavior our presented research showed that an acute administration receptor agonist oleamide (5-20 mg/kg) had no antagonist AM 251 (0.25-3 anxiogenic EPM test In turn, WIN55,212-2 at dose 1 mg/kg anxiolytic effect observed What interest both (JWH 133 doses 2 (AM 630 0.5-2 Moreover, revealed only inhibitor URB 597 (0.3 effect, while JZL 184 (at any (2-40 mg/kg)) after injection anxiety mice, confirmed clearly significant involvement responses. particular, able elicit promising effects. The body instinctively responds to external stimuli by increasing energy metabolism and initiating immune responses upon receiving stress signals. Corticosterone (CORT), a glucocorticoid (GC) that regulates secretion along the hypothalamic-pituitary-adrenal (HPA) axis, mediates neurotransmission humoral regulation. CB1 allosteric modulators such as synthetic cannabinoids are promising therapeutic agents. Among the various receptor modulators, indole-2-carboxamides, and N, N'-diphenyl urea derivatives continuously explored for their potency selectivity towards receptor. In present computational work, QSAR models were generated using Drug Theoretics Chemoinformatics (DTC tools) tools to analyze influence of molecular features these (one hundred fourteen compounds) on activity. A statistically significant three-parameter model (SPMin2_Bhm, maxdO, minssN) was derived that displayed R2 Q2 values 0.66 0.62, respectively. SPMin2_Bhm maxdO negatively correlated with activity, whereas minssN has a positive connection. seven-parameter (maxdO, minsssN, RDF55v, VE3_D, minHBint10, SpMin5_Bhs, CrippenLogP) also obtained 0.76 0.70, The findings might assist in design development novel based structures indole-2-carboxamides

Substance use disorders (SUDs) and anxiety (ADs) are highly comorbid, a co-occurrence linked to worse clinical outcomes than either condition alone. While the neurobiological mechanisms involved in SUDs intensively studied separately, underlying their comorbidity remain an emerging area of interest. This narrative review explores processes this comorbidity, using Research Domain Criteria (RDoC) framework map disruptions positive valence, negative cognitive systems across three stages addiction cycle: binge/intoxication, withdrawal/negative affect, preoccupation/anticipation. Anxiety substance play reciprocal role at each stage addiction, marked by significant psychosocial impairment dysregulation brain. A more thorough understanding neural underpinnings comorbid will contribute tailored effective therapeutic interventions assessments.
