Research findings indicate that programmed cell death (PCD) plays a pivotal role in the pathophysiology of spinal cord injury (SCI), and recently discovered form death, disulfidptosis, has emerged as novel phenomenon. However, characterization disulfidptosis-related genes SCI remains insufficiently explored. We retrieved SCI-related data from Gene Expression Omnibus (GEO) database identified three key associated with disulfidptosis human (CAPZB, SLC3A2, TLN1), whose mediated signaling pathways are closely intertwined SCI. Subsequent functional enrichment analysis suggested these may regulate multiple exert corresponding roles pathology. Moreover, we predicted potential targeted drugs for along their transcription factors constructed an intricate regulatory network. CIBERSORT revealed CAPZB, TLN1 might be implicated modulating changes within immune microenvironment individuals Our study provides compelling evidence confirming significant involvement following while offering valuable insights into its underlying pathological mechanisms.

The yak, a unique inhabitant of low-oxygen environments, exhibits brain adaptability to hypoxic conditions. However, the impact hypoxia on yak proteomics and expression HIF2α/BNIP3L signaling pathway remains unexplored. This study utilized TMT-based analysis identify differentially expressed proteins (DEPs) in cerebral cortexes 9-month-old yaks at high (n = 3) low altitudes. Additionally, qRT-PCR, Western blot, immunohistochemistry, immunofluorescence were used analyze HIF2α, BNIP3L, Beclin1, LC3-II, cleaved caspase-3 various regions from both altitude groups. KEGG revealed that DEPs mainly concentrated synthesis metabolism, DNA replication, repair pathways. Specifically, autophagy attracted our attention due its absence other animals. LC3-II increased significantly. Furthermore, results qRT-PCR blot showed that, same altitude, mRNA protein levels Beclin1 hippocampi significantly higher than those thalami, medulla oblongatae, cerebella (p < 0.05), while was not different among > 0.05). within region, high-altitude low-altitude yaks. Moreover, there no difference between Immunohistochemistry HIF2α-positive nucleus cytoplasm neurons, cytoplasm. caspase-3, NeuN co-located neurons cortex, hippocampus, thalamus, oblongata, cerebellum, respectively. offers complete characterization cortex proteome indicates these are more resistant hypoxia. The yak (Bos grunniens), the only large domesticated species endemic to Qinghai–Tibet Plateau, is a vital resource for local livelihoods and regional economic sustainability. However, breeding faces significant challenges, including limited understanding of genetic architecture underlying quantitative traits, inadequate advanced strategies, sterility hybrid offspring from yak–cattle crosses. These constraints have hindered progress in key production traits. To address these issues, integrating modern genomic tools into programs imperative. This review explores application potential molecular marker-assisted selection (MAS) prediction (GP) improvement. We systematically evaluate critical components pipelines, including: (1) phenotypic trait assessment, (2) sample collection (3) reference population design, (4) high-throughput genotyping (via genome sequencing SNP arrays), (5) predictive model development, (6) heritability estimation. By synthesizing current advances methodologies, this work aims provide framework leveraging technologies enhance efficiency, preserve diversity, accelerate gains populations.

Yaks are crucial genetic resources in the Tibetan Plateau and surrounding regions. Throughout long process of domestication, natural artificial selection pressures have enabled yaks to demonstrate adaptive characteristics environment terms physiological structure molecules, but no systematic cell analysis has been carried out on this phenomenon yaks. Here, population diversity yak were studied by WGRS, genes related adaptability excavated. Combined with scRNA-seq method, transcription map lung tissue skin was constructed, which provided a new comprehensive insight into adaptability. The showed that there obvious differentiation between TZ _ other seven populations, while significant exchange PL SB at high altitude. WGRS revealed gene HIF1A altitude adaptation expressed various types, EPAS1 predominantly epithelial endothelial cells tissue. Endothelial play critical role hypoxia-adapted VEGF signaling, correlates closely expression KDR VEGFA monocytes. Furthermore, signal High vs Low yak, 19.8 % overlapped screened scRNA-seq, including coat color such as RORA, BNC2, KIT. Notably, BNC2 is associated melanin deposition shows levels HS cells. Additionally, GRN melanocytes SORT1 IRS an important communication IRS. These findings offer insights polymorphism provide valuable reference for future research high-altitude mammals, potentially even human genetics.
