Knee osteoarthritis (KOA) is a degenerative joint disease characterized by the progressive deterioration of cartilage and synovial inflammation. A critical mechanism in pathogenesis KOA impaired efferocytosis tissue. The present study aimed to identify validate key efferocytosis-related genes (EFRGs) tissue using comprehensive bioinformatics machine learning approaches. We integrated three datasets (GSE55235, GSE55457, GSE12021) from Gene Expression Omnibus database screen differentially expressed (DEGs) associated with performed weighted gene co-expression network analysis. Subsequently, we utilized univariate logistic regression analysis, least absolute shrinkage selection operator regression, support vector machine, random forest algorithms further refine these genes. results were then inputted into multivariate analysis construct diagnostic nomogram. Public quantitative real-time PCR experiments employed for validation. Additionally, immune infiltration was conducted CIBERSORT combined datasets. Analysis intersection between DEGs EFRGs identified 12 KOA-related DEGs. Further refinement through revealed UCP2, CX3CR1, CEBPB as hub Immune demonstrated significant correlations cell components expression levels Validation independent experimental approaches confirmed robustness findings. This successfully (UCP2, CEBPB) alterations KOA, demonstrating high potential close associations efferocytosis. These targets may modulate processes, offering novel therapeutic avenues intervention.

This study aims to explore the relationship between preoperative and postoperative pain in patients with knee osteoarthritis (KOA) undergoing total arthroplasty (TKA) unicompartmental (UKR), assess mediating roles of central peripheral sensitization. prospective cohort enrolled a 105 eligible KOA patients. The Visual Analog Scale (VAS) was used levels. Multivariate linear regression analysis Bootstrap mediation were conducted investigate impact intensity on pain. revealed significant positive correlation VAS scores during movement 3 days postoperatively (β = 0.4018, p 0.0017), as well 6 months 0.735, 0.004). Central sensitization 0.0446, < 0.0001) 0.0333, 0.0015) exhibited effects Additionally, (B 1.107, 0.014, OR 3.027), age 0.207, 0.036, 1.229), duration arthritis 0.190, 0.013, 1.210) predictors analgesic demand. demonstrates that is closely related TKA UKR, playing this process. In particular, management crucial for alleviating improving patients' quality life. Therefore, when formulating individualized strategies, sensitivity should be comprehensively considered. Future research necessary develop evaluate innovative control methods integrating mechanisms optimize rehabilitation long-term health outcomes.
