Plasma-activated water (PAW), generated by cold plasma, is emerging as a potential treatment for colon cancer. This study focused on its anticancer effects against HCT-116 cancer cells, emphasizing the role of pH and conductivity variations due to plasma–fluid reactions. These changes suggest chemical transformation in PAW, leading increased acidity ion presence. The cytotoxic impact PAW cells was analyzed using methods like enzyme-linked immunosorbent assay microscopic evaluation. exhibited cytotoxicity but also affected normal posing challenge selectivity. An 18 h exposure duration identified balance between cell eradication preservation. Observed morphological indicated apoptotic characteristics PAW-treated hinting at mechanisms death. PAW-induced reactive oxygen species release mirrored cellular stress, with early markers, DNA fragmentation, heat shock proteins (HSPs) signifying complex responses. findings that can trigger apoptosis stress pathways cells. However, further studies are necessary therapy.

Background Acquired resistance to 5-fluorouracil (5-FU) frequently results in chemotherapy failure and disease recurrence advanced colorectal cancer (CRC) patients. Research has demonstrated that dysregulation of long non-coding RNAs (lncRNAs) mediates the development cancerous cells. The present study aims identify key lncRNAs associated with 5-FU CRC using bioinformatic experimental validation approaches. Methods Gene Expression Omnibus (GEO) dataset GSE119481, which contains miRNA expression profiles parental HCT116 cell line (HCT116/P) its in-vitro established 5-FU-resistant sub-cell (HCT116/FUR), was downloaded. Firstly, differentially expressed microRNAs (DEmiRNAs) between cells were identified. LncRNAs mRNAs then predicted online databases. Ontology (GO) Kyoto Encyclopedia Genes Genomes (KEGG) enrichment analyses performed uncover relevant biological mechanisms pathways. Networks integrating lncRNAs, miRNAs, interactions constructed, topological used resistance. An model HCT116/FUR developed by exposing HCT116/P increasing concentrations 5-FU. Finally, real-time quantitative PCR (RT-qPCR) on total RNA extracted from validate in-silico predictions lncRNAs. Results A 32 DEmiRNAs Enrichment analysis these mainly enriched several hallmark pathways regulate growth, cycle, survival, inflammation, immune response, apoptosis. predictive identified 237 unique 123 interacting DEmiRNAs. pathway indicated most genes cellular response starvation, protein polyubiquitination, chromatin remodeling, negative regulation gene expression. Topological lncRNA-miRNA-mRNA network highlighted nuclear abundant transcript 1 (NEAT1), metastasis-associated lung adenocarcinoma (MALAT1), Opa 5 antisense (OIP5-AS1) as central Experimental RT-qPCR confirmed levels NEAT1 MALAT1 significantly increased compared However, no significant difference observed OIP5-AS1 level two Conclusion Our findings specifically highlight contributors CRC. These are promising biomarkers for diagnosing predicting outcomes
