Abstract Since the onset of HIV epidemic, assessing CD4 + T-cells has become a routine procedure for evaluating immune deficiency, with flow cytometry established as gold standard. Over time, various strategies and platforms have been introduced to improve cell enumeration, aiming enhance performance diagnostic devices bring service closer patients. These advancements are particularly critical low-resource settings point-of-care applications, where excellent is hindered by its unsuitability in such environments. This work presents an innovative electrochemical microfluidic device that, further development, could be applied management low resource settings. The setup integrates sensor within PDMS structure, allowing on-chip electrode functionalization detection. Using impedance spectroscopy, biosensor demonstrates linear detection range from 1.25 × 10 5 2 6 cells/mL, limit 1.41 cells/mL cells isolated blood samples, aligning clinical ranges both healthy shows specificity towards negligible response monocytes, neutrophils, bovine serum albumin. Its integration chip fabrication detection, compact size, minimal manual handling, ease fabrication, capability, potential multiplexing together make advantageous use settings, standing out among other described literature. study also investigates Dean Flow Fractionation (DFF) separation.
