Industrial waste management is a growing concern, and the valorization of by-products through circular economy approaches represents sustainable solution. In this context, dextran–grape conjugates (PLG–GRAPE) were obtained via grafting reaction grape pomace extract dextran under aqueous conditions. To compare properties polymeric graft with those free extract, total polyphenol content was assessed using Folin–Ciocalteu assay, along stability diffusion studies. addition, in vitro safety evaluations, including Neutral Red Uptake, h-CLAT, skin irritation tests performed to assess biocompatibility. evaluate antioxidant, anti-inflammatory, anti-aging PLG–GRAPE, efficacy assays on keratinocyte fibroblast cell lines full-thickness reconstructed human tissues exposed damaging agents, such as UV radiation pollutants. The results showed that technology preserved phenolic antioxidant activity while improving properties. As demonstrated by studies, favorable biocompatibility profile observed, addition significant capacity reduce oxidative stress inflammation aged cells, thus, attenuating cellular aging senescence. conclusion, study suggests PLG–GRAPE has potential bioactive ingredient for cosmetic dermatological applications, offering effective approach utilizing industrial products.

Background: Grape pomace (GP), a by-product of winemaking, is rich source bioactive polyphenols known for their antioxidant and anti-inflammatory properties. While the cardiovascular benefits red grape have received significant scientific attention, therapeutic potential white remains largely unexplored, particularly in glucocorticoid-induced hypertension. Given rising prevalence hypertension oxidative-inflammatory mechanisms underlying its progression, this study investigates effects GP on blood pressure regulation, oxidative stress, pro-inflammatory cytokine expression an experimental model dexamethasone (DEXA)-induced (HTN). By focusing GP, research addresses gap current knowledge proposes novel, sustainable approach to managing through valorising winemaking by-products. Methods: The first concentration used, GP1, was 795 mg polyphenols/kg bw, while second concentration, GP2, 397.5 bw. Results: White extract DEXA_GP1 group had reduced systolic diastolic pressure. with higher content (GP1) prevented elevation serum levels total stress (TOS), malondialdehyde (MDA), index (OSI), lower (GP2) slightly MDA. Both concentrations increased NO Total Thiols, significantly (p < 0.05) than treated lisinopril. tumour necrosis factor-alpha (TNF-α) all groups where HTN induced. doses TNF-α. Heart tissue studied cytokines (TNF-α, interleukin (IL)-1β, IL-6 were not influenced > by either induction or treatment administered. Conclusions: These findings suggest that may serve as promising nutraceutical intervention management, conditions associated stress.

This study investigates the anti-inflammatory properties polyphenolic fraction derived from GP, obtained by ultrasound-microwave hybrid extraction and purified using ion-exchange chromatography. In inflammation model, mice were divided into six groups: intact, carrageenan, indomethacin, three GP treatment groups. Paw edema was induced subplantar injection administered intraperitoneally at doses 10, 20, 40 mg/kg. The effect evaluated measuring paw volume, expression inflammatory markers: cyclooxygenase-2 (COX-2), myeloperoxidase (MPO), cytokines (IL-1β IL-6), along lipid peroxidation levels. significantly reduced levels COX-2, MPO, in dose-dependent manner effect, highest dose showing greatest reduction. Additionally, also decreased 10 20 These findings suggest that combined amberlite purification proved to be effective obtaining polyphenolic-rich GP. Thus, may serve as natural antioxidant agent for treating oxidative stress-related diseases.
