In this work, and 5 analogues (BPS, BPAP, BPAF, BPFL BPC) have been studied classical SH-SY5Y the alternative 3D vitro models after 24 96 h exposure. Abstract Bisphenol-A (BPA) is widely used in food packaging and household products, leading to daily human exposure potential health risks including metabolic diseases like type 2 diabetes mellitus (T2DM). Understanding BPA's mechanisms developing intervention strategies urgent. Centella asiatica , a traditional herbal medicine containing pentacyclic triterpenoids, shows promise due its antioxidant anti-inflammatory properties, utilized for centuries Ayurvedic therapy. We investigated the effect of (CA) ethanol extract on BPA-induced pancreatic islet toxicity male Swiss albino mice. BPA administration (10 100 μg/kg body weight, twice daily) 21 days caused glucose homeostasis disturbances, insulin resistance, dysfunction, which were partially mitigated by CA supplementation (200 400 mg/kg weight). Additionally, heightened oxidative stress, elevated levels proinflammatory cytokines, loss mitochondrial membrane (MMP), abnormal cell cycle, increased apoptosis implicated detrimental impact endocrine pancreas effectively counteracted supplementation. In summary, demonstrated significant ability mitigate apoptosis, modulate redox homeostasis, alleviate inflammation, preserve MMP, regulate cycle. This study explores the neuroprotective effects of resveratrol (Resv) against tri-o-cresyl phosphate (TOCP)-induced neurotoxicity in spinal cord adult hens. It is well documented that TOCP exposure causes significant neurodegeneration via mechanisms involve endoplasmic reticulum (ER) stress and impaired autophagy. In this experiment, hens were assigned to one four groups: Control, Resv, TOCP, + Resv. The tissues examined through transmission electron microscopy, hematoxylin eosin (HE) staining, Nissl Western blotting evaluate key proteins associated with ER Additionally, RT-qPCR immunofluorescence employed measure sirtuin1 (SIRT1) expression. findings revealed induced severe ultrastructural damage, including disrupted myelin sheaths, dilated ER, extensive neurodegeneration, as confirmed by histological evaluations. expression levels GRP78, p-PERK, p-eIF2α, ATF4, CHOP, Beclin-1, P62, LC3-II also significantly elevated TOCP. However, Resv treatment markedly attenuated these pathological changes reducing stress, restoring autophagic flux, upregulating SIRT1 expression, preserving integrity. These results indicate can effectively counteract TOCP-induced modulating autophagy, underscoring its potential a therapeutic agent for neuroprotection.
