With changing lifestyles, non-alcoholic fatty liver disease (NAFLD) has become the most prevalent worldwide. A substantial increase in incidence, mortality, and associated burden of NAFLD-related advanced is expected. Currently, initial diagnosis NAFLD still based on ultrasound there no approved treatment method. Lipid-lowering drugs, vitamin supplementation, lifestyle improvement treatments are commonly used clinical practice. However, lipid-lowering drugs can produce poor patient compliance specific adverse effects. Therefore, exploration bio-diagnostic markers active lead compounds for development innovative urgently needed. More more studies have reported anti-NAFLD effects mechanisms natural products (NPs), which an important source new drug to treat due their high activity low side At present, berberine silymarin been by US FDA enter phase IV studies, demonstrating potential NPs against NAFLD. Studies found that regulation lipid metabolism, insulin resistance, oxidative stress, inflammation-related pathways may play roles process. continuous updating technical means scientific theories, in-depth research targets provide possibilities find drugs. Abstract Nonalcoholic fatty liver disease (NAFLD) has become one of the most common chronic diseases. O‐Linked attachment beta‐N‐acetylglucosamine ( O ‐GlcNAc) are ubiquitous post‐translational modifications proteins as “nutrient sensors” and “stress receptors” in body that involved maintaining normal cellular physiological functions. Increased levels ‐GlcNAcylation have been found samples patients with NAFLD nonalcoholic steatohepatitis. However, role development pathogenesis remains unclear. Here, we sought to determine specific NAFLD. In this study, results demonstrated inhibition ‐GlcNAc transferase (OGT) led decreased expression lipid synthesis genes vitro. addition, showed acid synthase (FASN) was positively correlated levels. Immunoprecipitation pulldown assays confirmed interaction between FASN OGT at serine 1483 FASN, inhibit K48‐linked ubiquitination degradation thereby promoting hepatic accumulation Administration inhibitor OSMI‐1 ob/ob mice accumulation, further confirming our vitro experimental results. Finally, used liver‐specific Ogt gene knockout fed a high‐fat diet elucidate mechanism on knockdown accumulation. conclusion, findings show inhibiting S1483 site promotes leads liver. Abstract A prevalent liver condition called non-alcoholic fatty disease (NAFLD) may progress into steatohepatitis (NASH) and cause life-threatening complications like cirrhosis cancer. The development progression of NAFLD has been linked to the make-up functioning gut microflora. This article reviews clinical studies reported investigate connection between changes in microbiota metabolic markers patients. According study findings, dysbiosis microflora were observed patients, which are manifested by variations proportions particular bacterial species. Glucose and lipid metabolism dysregulation in skeletal muscle contributes to the development of metabolic disorders. The efficacy fucoxanthin alleviating disorders remains poorly understood. In this study, we systematically investigated impact on mitigating deposition insulin resistance employing palmitic acid-induced C2C12 cells ob/ob mice. Fucoxanthin significantly alleviated PA-induced resistance. addition, prominently upregulated expression metabolism-related genes (Pparα Cpt-1), promoting fatty acid β-oxidation metabolism. Additionally, increased Pgc-1α Tfam, elevated mtDNA/nDNA ratio, reduced ROS levels. Further, identified pyruvate kinase isozyme 1 (PKM1) as a high-affinity protein for by drug affinity-responsive target stability LC–MS confirmed their robust interaction CETSA, microscale thermophoresis, circular dichroism. Supplementation with pyruvate, product PKM1, attenuated beneficial effects Mechanistically, glucose glycolysis rate enhanced mitochondrial biosynthesis through inhibiting PKM1 activity, thereby stress. The Bouguer-Lambert-Beer (BLB) law serves as the fundamental basis for spectrophotometric determination of pigment content in microalgae. Although it has been observed that applicability BLB is compromised by light scattering effect microalgae suspensions, in-depth research concerning relationship between and accuracy remains scarce. We hypothesized (1) precision using would diminish with increasing nonlinearity absorbance, (2) employing modified version (mBLB) yield superior performance. To assess our hypotheses, we cultivated Phaeodactylum tricornutum under varying illumination conditions nitrogen supplies controlled indoor experiments, resulting suspensions diverse contents. Nonalcoholic steatohepatitis (NASH) is a progressive form of nonalcoholic fatty liver disease (NAFLD) associated with metabolic syndrome and increased cardiovascular risk. Resmetirom, novel liver-directed selective thyroid hormone receptor-β (THR-β) agonist, has shown promise in addressing both hepatic systemic lipid metabolism. This systematic review meta-analysis aimed to evaluate the efficacy resmetirom improving cholesterol levels NASH patients. A literature search was conducted across multiple databases including PubMed, Embase, Cochrane Library, ClinicalTrials.gov, identifying three randomized controlled trials for inclusion. The revealed that significantly reduced low-density lipoprotein (LDL-C) compared placebo (MD: -23.62; 95% CI: -37.32 -9.93; p < 0.001). Similarly, triglyceride (TG) showed significant reduction group -33.86; -47.79 -19.92; Importantly, there no difference risk serious adverse events between groups (RR: 1.09; 0.73 1.63; = 0.67). These findings suggest effectively improves profiles patients without compromising safety. However, analysis limited by small number studies, all from same research group, high heterogeneity results. Future should include more diverse longer follow-up periods, cost-effectiveness evaluations. Despite these limitations, shows as potential treatment managing dyslipidemia patients, potentially influencing future guidelines health this population.
