Obesity
is
defined
as
chronic,
low-grade
inflammation
within
specific
tissues.
Given
the
escalating
prevalence
of
obesity
among
individuals
all
ages,

epidemic
proportions,
posing

important
public
health
challenge.
Despite
significant
advancements
in
treating
obesity,
conventional
approaches
remain
largely
ineffective
or
involve
severe
side
effects,
thus
underscoring
pressing
need
to
explore
and
develop
treatment
approaches.
Targeted
local
immunomodulation
using
nanoparticles
(NPs)
can
influence
fat
production
utilization
processes.
Statins,
known
for
their
anti-inflammatory
properties,
show
potential
mitigating
obesity-related
inflammation.
A
localized
delivery
option
offers
several
advantages
over
oral
parenteral
methods.
Here,
we
developed
simvastatin
(Sim)
encapsulated
PLGA
NPs
(Sim-NP)
Sim
adipose
tissues
(ATs)
treat
obesity.
In
vitro
experiments
revealed
strong
effects
Sim-NPs,
which
resulted
enhanced
modulation
macrophage
(MΦ)
polarization
induction

browning.

The Association between Statin Use and Reduced Migraine Likelihood: A Comprehensive Analysis of Migraine Subtypes and Statin Types in a Nationwide Korean Cohort

Despite
growing
interest
in
the
preventive
effects
of
statins,
as
lipid-lowering
agents,
on
migraine
attacks,
comprehensive
nationwide
studies
comparing
likelihood
between
statin
users
and
controls
are
lacking.
Our
nested
case–control
study
within
Korean
National
Health
Insurance
Service-Health
Screening
Cohort
(2002–2019)
investigated
this
association
using
38,957
patients
155,828
controls,
considering
subtypes
(with/without
aura)
types
(lipophilic
vs.
hydrophilic).
Using
propensity
score
matching
adjusting
for
confounders,
use
was
linked
to
reduced
overall
(odds
ratio
(OR)
0.93),
particularly
migraines
with
aura
(OR
0.75)
without
0.94).
Lipophilic
statins
were
effective
both
subtypes,
while
hydrophilic
mainly
aura.
Subgroup
analyses
showed
consistent
benefits
across
demographics,
but
varied
effectiveness
based
weight,
smoking,
alcohol
use,
hemoglobin
levels,
dyslipidemia
history.
In
summary,
cohort
suggests
that
may
reduce
among
adults
diverse
demographics
clinical
profiles,
certain
lifestyle
comorbidity
factors
underscores
importance
individual
patient
profiles
when
assessing
potential
therapy
prevention.

Down
syndrome
(DS)
is
a
complex
chromosomal
disorder
considered
as
genetically
determined
form
of
Alzheimer’s
disease
(AD).
Maintenance
brain
cholesterol
homeostasis
essential
for
functioning
and
development,
its
dysregulation
associated
with
AD
neuroinflammation
oxidative
damage.
Brain
imbalances
also
likely
occur
in
DS,
concurring
the
precocious
AD-like
neurodegeneration.
In
this
pilot
study,
we
analyzed,
Ts2Cje
(Ts2)
mouse
model
expression
genes
encoding
key
enzymes
involved
metabolism
levels
main
precursors
products
(i.e.,
oxysterols).
The
results
showed,
Ts2
mice
compared
to
euploid
mice,
downregulation
transcription
3-hydroxy-3-methylglutaryl-CoA
reductase
24-dehydrocholesterol
reductase,
latter
originally
recognized
an
indicator
AD,
consequent
reduction
total
levels.
Moreover,
responsible
oxidation
amounts
resulting
oxysterols
were
modified
brains,
autoxidation
increased,
suggesting
exacerbation
cerebral
stress.
We
observed
enhanced
inflammatory
response
underlined
by
upregulation
α-interferon
interleukin-6,
two
cytokines
whose
synthesis
increased
brains
patients.
Overall,
these
suggest
that
DS
share
cycle
derangements
altered
oxysterol
levels,
which
may
contribute
events
both
diseases.
