Nanotechnology holds significant ameliorative potential against neurodegenerative diseases, as it can protect the therapeutic substance and allow for its sustained release. In this study, reducing capping agents of Urtica dioica (UD), Matricaria chamomilla (MC), Murraya koenigii (MK) extracts were used to synthesize bio-mediated zinc oxide nanoparticles (ZnO-NPs) bacteria (Staphylococcus aureus Escherichia coli) rotenone-induced toxicities in D. melanogaster first time. Their optical structural properties analyzed via FT-IR, DLS, XRD, EDS, SEM, UV–Vis, zeta potential. The antioxidant antimicrobial fabricated ZnO-NPs evaluated employing cell-free models (DPPH ABTS) well diffusion method, respectively. Rotenone (500 µM) was administered Drosophila third instar larvae freshly emerged flies 24–120 h, either alone or combination with plant (UD, MC, an MK) their biogenic ZnO-NPs. A comparative study on protective effects synthesized NPs undertaken neurotoxic, cytotoxic, behavioral alterations using acetylcholinesterase inhibition assay, dye exclusion test, locomotor parameters. findings revealed that among plant-derived ZnO-NPs, MK-ZnO exhibit strong activities, followed by UD-ZnO MC-ZnO NPs. regard, ethno-nano medicinal uses mimic similar suppressing oxidative stress restoring biochemical parameters (AchE proteotoxicity activity) lower cellular toxicity. These suggest green-engineered have significantly enhance outcomes, promise effective therapies neurodegeneration, could be a great alternative clinical development.
