Background Cancer stem cells (CSCs) are able to survive after cancer therapies, resulting in tumor progression and recurrence, as is seen colorectal cancer. Therapies targeting CSCs regarded novel promising strategies for efficiently eradicating tumors. Berberine, an isoquinoline alkaloid extracted from the Chinese herbal medicine Coptis chinensis, was found have antitumor activities against cancer, without knowing whether it exerts inhibitory effects on potential mechanisms. Methods In this study, we examined roles of Berberine derived HCT116 HT29 by culturing serum-free medium. We also m 6 A methylation via regulating fat mass obesity-associated protein (FTO), downregulating β-catenin. Results tumorigenicity, growth, stemness stem-like cells. The regulatory effect N6-methyladenosine (m A), abundant mRNA modification, examined. treatment decreased cell proliferation decreasing cyclin D1 increasing p27 p21 subsequently induced cycle arrest at G 1 /G 0 phase. colony formation apoptosis. transcriptionally increased FTO thus methylation, which reversed both knockdown addition inhibitor FB23-2. FTO-related decreases CSCs. chemosensitivity promoted chemotherapy agent-induced Moreover, that β-catenin, a negative regulator FTO. Conclusions Our observation effectively β-catenin suggests role modulating malignant behaviors

Abstract Berberine (BBR) is a potential plant metabolite and has remarkable anticancer properties. Many kinds of research are being focused on the cytotoxic activity berberine in vitro vivo studies. A variety molecular targets which lead to effect ranges from p‐53 activation, Cyclin B expression for arresting cell cycles; protein kinase (AKT), MAP IKB antiproliferative activity; beclin‐1 involved autophagy; reduced MMP‐9 MMP‐2 inhibition invasion metastasis etc. also interferes with transcription factor‐1 (AP‐1) responsible oncogenes neoplastic transformation cell. It leads various enzymes directly or indirectly carcinogenesis like N acetyl transferase, Cyclo‐oxygenase‐2, Telomerase Topoisomerase. In addition these actions, plays role in, regulation reactive oxygen species inflammatory cytokines preventing cancer formation. properties demonstrated due interaction micro‐RNA. The summarized information presented this review article may help researchers, scientists/industry persons use as promising candidate against cancer.
