Atherosclerosis (AS) seriously affects human health. The role of microRNAs (miRNAs) in the pathogenesis and progression AS has become a focus research. Our goal was to identify biological effect differentially expressed miRNAs (DE-miRNAs) AS.To analyze genes (DEGs), including mRNAs (DE-mRNAs) DE-miRNAs, by using Gene Expression Omnibus (GEO) database limma package. DEGs protein-protein interaction (PPI) network functional enrichment analysis were constructed search tool for retrieval interacting genes/proteins (STRING) database, Cytoscape software plugin "ClueGO2.5.6". We established coexpression dysregulated predict function miRWalk Pearson correlation coefficient (PCC) analysis. Cellular experiments used validate results bioinformatics.First, 69 common obtained from datasets GSE43292 GSE97210 package R. Next, DEG PPI constructed. Functional showed that 11 pathways significantly enriched, such as positive regulation monocyte chemotaxis. Seven DE-miRNAs GSE99685 dataset DE-mRNAs predicted through database. miRNA-mRNA suggested miR-148a-3p targeted contactin 4 (CNTN4). Quantitative real-time polymerase chain reaction (qRT-PCR) assay indicated downregulated CNTN4 upregulated THP-1 + phorbol 12-myristate 13-acetate (PMA) oxidized low-density lipoprotein (oxLDL) group compared with PMA group. qRT-PCR, flow cytometry, enzyme-linked immunosorbent (ELISA) found inhibited expression CNTN4, cell apoptosis, interleukin-6 (IL-6) tumor necrosis factor-alpha (TNF-α) concentrations oxLDL-induced macrophages.
