Vanadium is a transition metal with important industrial, technological, biological, and biomedical applications widespread in the environment living beings. Repurposing drugs by uncovering new indications for approved accelerates the process of establishing treatments and reduces high costs drug discovery development. Metal complexes with clinically allow further opportunities in cancer therapy—many vanadium compounds have previously shown antitumor effects, which makes a suitable metal to complex therapeutic drugs, potentially improving their efficacy treatment. In this review, covering last 25 years research field, we identified non-oncology-approved as ligands obtain different complexes. Metformin-decavanadate, vanadium-bisphosphonates, vanadyl(IV) non-steroidal anti-inflammatory cetirizine imidazole-based oxidovanadium(IV) complexes, each has parent known medicinal properties indications, all showed potential novel anticancer treatments. Nevertheless, precise mechanisms action these against are still not fully understood.

Abstract Giant heterometallic polyoxometalate (POM) clusters with precise atom structures, flexibly adjustable and abundant active sites are promising for constructing functional nanodrugs. However, current POM drugs almost vacant in orthotopic brain tumor therapy due to the inability effectively penetrate blood–brain barrier (BBB) low drug activity. Here, we designed largest (3.0 nm × 6.0 nm) transition‐metal–lanthanide co‐encapsulated cluster {[Ce 10 Ag 6 (DMEA)(H 2 O) 27 W 22 O 70 ][B‐ α ‐TeW 9 33 ] } 88− featuring 238 metal centers via synergistic coordination between two geometry‐unrestricted Ce 3+ + linkers tungsten‐oxo fragments. This was combined brain‐targeted peptide prepare a nanodrug that could efficiently traverse BBB target glioma cells. The specifically activate reactive oxygen species regulate apoptosis pathway of cells half‐maximal inhibitory concentration (5.66 μM). As first drug, it prolongs survival glioma‐bearing mice.
