OTR signaling promotes social approach or facilitates avoidance, depending on the brain region involved. The lateral septum plays a critical role in regulating interactions and memory. Dysfunction in social interactions is a core symptom of autism spectrum disorder (ASD). Nevertheless, the neural mechanisms underlying deficits ASD are poorly understood. By integrating electrophysiological, vivo fiber photometry, viral-mediated tracing, optogenetic and pharmacological stimulation, we show reduced intrinsic excitability hypoactivity SOM interneurons medial prefrontal cortex (mPFC) Magel2-deficient mice, an established model, were required to defects. Anxiety is a prevalent mental illness known for its high incidence, comorbidity, and tendency to recur, posing significant societal individual burdens. Studies have highlighted Interleukin-19 (IL-19) as having potential relevance in neuropsychiatric disorders. Our previous research revealed that IL-19 overexpression colonies exacerbated anxiety-related behaviors induced by dextran sodium sulfate/stress. However, the precise role molecular mechanisms of anxiety regulation remain uncertain. In this study, we initiated an acute restraint stress (ARS)-induced anxious mouse model identified heightened expression IL-20Rα medial prefrontal cortex (mPFC) ARS mice. Notably, were predominantly present excitatory pyramidal neurons mPFC under both basal conditions. Utilizing adeno-associated virus (AAV) strategy, demonstrated elevated susceptibility.
